大蒜素對(duì)兔膝骨關(guān)節(jié)炎作用的實(shí)驗(yàn)研究

【摘要】:[目的]探討大蒜素對(duì)兔膝骨關(guān)節(jié)炎的作用。[方法]將30只家兔隨機(jī)分為空白對(duì)照組、模型對(duì)照組和大蒜素組,每組10只;除空白對(duì)照組外其余2組按改良Hulth法建立模型,術(shù)后開始灌服藥物,大蒜素組(大蒜素,128 mg/kg/d)、空白對(duì)照組和模型對(duì)照組(等量生理鹽水20 ml),連用8周。術(shù)后8周處死所有家兔取右膝股骨髁大體觀察;蘇木精-伊紅(HE)和番紅O(SA-O)染色對(duì)股骨髁關(guān)節(jié)軟骨行Mankin評(píng)分;采用q RTPCR法檢測(cè)股骨內(nèi)側(cè)髁負(fù)重區(qū)軟骨組織interleukin-1β(白細(xì)胞介素-1β,IL-1β)、matrix metalloproteinase-3(基質(zhì)金屬蛋白酶-3,MMP-3)和tissue inhibitor of metalloproteases-1(基質(zhì)金屬蛋白酶組織抑制因子-1,TIMP-1)的m RNA相對(duì)表達(dá)量。[結(jié)果]三組之間比較,空白對(duì)照組的Mankin評(píng)分最低,IL-1β、MMP-3和TIMP-1 m RNA表達(dá)量最低,模型對(duì)照組和大蒜素組,分別與空白對(duì)照組比較,上述指標(biāo)差異均有統(tǒng)計(jì)學(xué)意義(P0.05);與模型對(duì)照組比較,大蒜素組Mankin評(píng)分降低,IL-1β、MMP-3、TIMP-1 m RNA表達(dá)量降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。[結(jié)論]大蒜素能夠改善兔膝骨關(guān)節(jié)炎Mankin評(píng)分并能夠下調(diào)股骨髁關(guān)節(jié)軟骨中IL-1βm RNA、MMP-3m RNA表達(dá),上調(diào)TIMP-1 m RNA表達(dá)量。
[Abstract]:[objective] to investigate the effect of allicin on knee osteoarthritis in rabbits. [methods] Thirty rabbits were randomly divided into blank control group, model control group and allicin group with 10 in each group. Allicin group (allicin 128mg / kg / d), blank control group and model control group (20 ml saline, 8 weeks). Eight weeks after operation, all the rabbits were sacrificed for gross observation of the femoral condyle of the right knee, and the Mankin scores of the femoral condylar cartilage were evaluated by hematoxylin eosin (HEH) and guanghong (OSA-O) staining. The expression of interleukin-1 尾 (IL-1 尾) matrix metalloproteinase-3 (MMP-3) and tissue inhibitor of metalloproteases-1 (TIMP-1) in cartilage of the medial condylar of femur were detected by Q RTPCR method. [results] the expression of IL-1 尾 -MMP-3 and TIMP-1 m RNA in the blank control group was the lowest, and that in the model control group and allicin group was the lowest, respectively, compared with the blank control group. Compared with the model control group, the Mankin score of allicin group was lower than that of the model control group, and the expression of IL-1 尾, MMP-3, TIMP-1 m RNA was significantly lower in allicin group than that in the model control group (P 0.05). [conclusion] allicin can improve the Mankin score of rabbit knee osteoarthritis, down-regulate the expression of IL-1 尾 -RNA-3m RNA and up-regulate the expression of TIMP-1 m RNA in femoral condylar articular cartilage.
