迷走神經(jīng)電刺激對(duì)腦外傷后昏迷大鼠前額葉皮質(zhì)去甲腎上腺素α1受體表達(dá)變化的影響

【摘要】:目的:探討迷走神經(jīng)電刺激(vagus nerve stimulation,VNS)對(duì)腦外傷(traumatic brain injury,TBI)后昏迷大鼠的促醒作用及其可能機(jī)制。方法:健康成年SD大鼠90只,隨機(jī)分為空白對(duì)照組、假刺激組和刺激組。TBI后昏迷大鼠給予VNS治療,觀察其行為學(xué)變化,并通過(guò)免疫組織化學(xué)和Western-blot技術(shù)檢測(cè)各實(shí)驗(yàn)組大鼠前額葉皮質(zhì)(prefrontal cortex,PFC)組織中去甲腎上腺素α1受體(α1R)含量。結(jié)果:假刺激組和刺激組大鼠各30只,觀察行為學(xué)變化發(fā)現(xiàn),刺激組中20只出現(xiàn)翻正反射,假刺激組僅8只出現(xiàn)翻正反射;將3個(gè)實(shí)驗(yàn)組中α1R含量進(jìn)行兩兩比較,發(fā)現(xiàn)刺激組α1R水平均顯著高于其他兩組(P0.05)。結(jié)論:VNS可改善TBI后昏迷大鼠的意識(shí)狀態(tài)水平,對(duì)促進(jìn)TBI后昏迷大鼠的覺(jué)醒有積極的治療作用,其機(jī)制可能與PFC去甲腎上腺素α1R水平上調(diào)有關(guān)。
[Abstract]:Objective: to investigate the effects of vagus nerve stimulation (VNS) on the wake-up of coma rats after traumatic brain injury-induced brain injury (TBI) and its possible mechanism. Methods: 90 healthy adult SD rats were randomly divided into control group, sham stimulation group and stimulative group. The rats in coma group were treated with VNS, and their behavioral changes were observed. The content of norepinephrine 偽 1 receptor (偽 1R) in prefrontal cortex of rats in each experimental group was determined by immunohistochemistry and Western-blot technique. Results: there were 30 rats in the sham stimulation group and 30 rats in the stimulation group. The behavioral changes showed that 20 rats in the stimulation group and 8 rats in the sham stimulation group had positive inversion reflex, and the contents of 偽 1R in the three experimental groups were compared in pairs. It was found that the level of 偽 1R in the stimulation group was significantly higher than that in the other two groups. ConclusionVNS can improve the level of consciousness in coma rats after TBI, and has a positive therapeutic effect on the arousal of coma rats after TBI. The mechanism may be related to the up-regulation of PFC norepinephrine 偽 1R level.
