【摘要】:[目的]通過(guò)觀察TGF-β1和α-SMA在人的損傷及正常半月板組織的表達(dá)情況,并進(jìn)行相關(guān)性分析,探討半月板損傷時(shí)兩者的變化情況。為進(jìn)一步研究半月板損傷愈合的機(jī)制提供實(shí)驗(yàn)依據(jù)。[方法]收集2014年3月至2014年12月52例確診為半月板損傷的半月板組織以及11例正常半月板組織,3度損傷半月板組織取自關(guān)節(jié)鏡手術(shù)中,急性期17例、慢性期16例;2度損傷半月板組織取自全膝關(guān)節(jié)置換手術(shù)中,慢性期19例,以11例正常半月板組織作為正常對(duì)照組。通過(guò)免疫組織化學(xué)染色方法對(duì)TGF-β1和α-SMA在各組中的表達(dá)情況進(jìn)行檢測(cè),并按損傷時(shí)間、損傷程度、年齡進(jìn)行分組比較,采用陽(yáng)性細(xì)胞百分?jǐn)?shù)進(jìn)行定量分析,統(tǒng)計(jì)的數(shù)據(jù)采用SPSS19.0軟件分析,并運(yùn)用方差分析各組差異,直線相關(guān)分析法分析TGF-β1和α-SMA表達(dá)的相關(guān)性。[結(jié)果]1.TGF-β1在3度損傷急性期組、3度損傷慢性期組陽(yáng)性細(xì)胞百分?jǐn)?shù)均高于正常組(p0.05);3度損傷急性期組陽(yáng)性細(xì)胞百分?jǐn)?shù)高于3度損傷慢性期組(p0.05)。2.α-SMA在3度損傷急性期組、3度損傷慢性期組陽(yáng)性細(xì)胞百分?jǐn)?shù)均高于正常組(p0.05);3度損傷急性期組陽(yáng)性細(xì)胞百分?jǐn)?shù)高于3度損傷慢性期組(p0.05)。3.TGF-β1在慢性期2度損傷組、慢性期3度損傷組陽(yáng)性細(xì)胞百分?jǐn)?shù)均高于正常組(p0.05);慢性期2度損傷組陽(yáng)性細(xì)胞百分?jǐn)?shù)高于慢性期3度損傷組(p0.05)。4.α-SMA在慢性期2度損傷組、慢性期3度損傷組陽(yáng)性細(xì)胞百分?jǐn)?shù)均高于正常組(p0.05);慢性期2度損傷組陽(yáng)性細(xì)胞百分?jǐn)?shù)高于慢性期3度損傷組(p0.05)。5.TGF-β1、α-SMA在損傷半月板中≥45歲組與45歲組的陽(yáng)性細(xì)胞百分?jǐn)?shù)差異均無(wú)統(tǒng)計(jì)學(xué)意義(p0.05)。6.在各實(shí)驗(yàn)組及正常對(duì)照組中,TGF-β1的表達(dá)與α-SMA的表達(dá)之間呈明顯正相關(guān)性,具有統(tǒng)計(jì)學(xué)意義(r值=0.971,p0.05)。[結(jié)論]1.TGF-β1和α-SMA在3度損傷中急性期的表達(dá)升高,隨著損傷時(shí)間的增加而表達(dá)降低。2.TGF-β1和α-SMA在慢性期損傷中,損傷程度越大而表達(dá)越低。3.在各實(shí)驗(yàn)組與正常對(duì)照組中,TGF-β1和α-SMA的表達(dá)之間存在正相關(guān)性。
[Abstract]:[objective] to observe the expression of TGF- 尾 1 and 偽 -SMA in human and normal meniscus tissues, and to investigate the changes of TGF- 尾 1 and 偽 -SMA in meniscus injury. To provide experimental basis for further study of meniscus healing mechanism. [methods] from March 2014 to December 2014, 52 cases of meniscus tissue diagnosed as meniscus injury and 11 cases of normal meniscus tissue were collected from arthroscopic surgery. The meniscus tissue of 16 patients with chronic meniscus injury was taken from the total knee replacement operation. In the chronic stage 19 cases were treated with normal meniscus tissue as the normal control group. The expression of TGF- 尾 1 and 偽 -SMA in each group was detected by immunohistochemical staining. The expression of TGF- 尾 1 and 偽 -SMA in each group were compared according to the time, degree and age of injury, and the percentage of positive cells was used to quantitatively analyze the expression of TGF- 尾 1 and 偽 -SMA in each group. The statistical data were analyzed by SPSS19.0 software. The correlation between TGF- 尾 1 and 偽 -SMA expression was analyzed by linear correlation analysis. [results] 1. The percentage of positive cells of TGF- 尾 1 in the acute phase of the third degree injury group was higher than that in the control group in the chronic phase of the third degree injury. The percentage of positive cells in the acute phase group was higher than that in the chronic phase group of the third degree injury group. 偽 -SMA was found in the acute phase of the third degree injury group. The percentage of positive cells in the chronic stage group was higher than that in the acute stage group in the normal group, and the percentage of the positive cells in the acute stage group was higher than that in the chronic stage group. 3. TGF- 尾 1 was higher in the second degree injury group in the chronic stage. The percentage of positive cells in the third degree chronic injury group was higher than that in the normal group, and the percentage of positive cells in the second degree chronic injury group was higher than that in the third degree chronic injury group, and 偽 -SMA was higher in the second degree chronic injury group. The percentage of positive cells in the third degree chronic injury group was higher than that in the normal group, and the percentage of positive cells in the second degree chronic injury group was higher than that in the chronic third degree injury group. 5.
