術(shù)后認(rèn)知功能障礙小鼠miR-146a表達(dá)的變化

【摘要】:目的觀察術(shù)后認(rèn)知功能障礙(postoperative cognitive dysfunction,POCD)小鼠術(shù)后血清、海馬及前額葉皮質(zhì)中miR-146a表達(dá)的變化。方法雄性C57BL/6小鼠50只,12~14周齡,體重25~30g,采用隨機(jī)數(shù)字表法將其分為兩組:對(duì)照組(C組)和麻醉手術(shù)組(AS組),每組25只。AS組在2.1%異氟醚吸入麻醉下行脛骨骨折開(kāi)放復(fù)位內(nèi)固定術(shù),C組不行麻醉和手術(shù)處理。于術(shù)前24h及術(shù)后6、12、24、48h每組分別取3只小鼠進(jìn)行摘眼球取血,處死后取海馬和前額葉皮質(zhì),采用RT-PCR法測(cè)小鼠血清、海馬和前額葉皮質(zhì)中miR-146a表達(dá)量。于術(shù)后第1、3、7天每組各取10只小鼠,進(jìn)行場(chǎng)景恐懼記憶實(shí)驗(yàn)和聲音提示恐懼記憶實(shí)驗(yàn),記錄僵直時(shí)間百分比。結(jié)果與C組比較,AS組場(chǎng)景恐懼記憶實(shí)驗(yàn)中僵直時(shí)間百分比明顯降低(P0.05),聲音提示恐懼記憶實(shí)驗(yàn)中兩組僵直時(shí)間百分比差異無(wú)統(tǒng)計(jì)學(xué)意義。在血清中,與C組比較,術(shù)后6、12、24、48h AS組miR-146a表達(dá)量明顯升高(P0.05);與術(shù)后12h比較,術(shù)后6、24、48hAS組miR-146a表達(dá)量明顯降低(P0.05)。在海馬中,與C組比較,術(shù)后6、12、24、48h AS組miR-146a表達(dá)量明顯升高(P0.05);與術(shù)后12h比較,術(shù)后6、48h AS組miR-146a表達(dá)量明顯降低(P0.05)。在前額葉皮質(zhì)中,與C組比較,AS組術(shù)后24、48h miR-146a表達(dá)量明顯升高(P0.05);與術(shù)后24h比較,術(shù)后48h AS組miR-146a表達(dá)量明顯升高(P0.05)。結(jié)論 POCD小鼠血清、海馬及前額葉皮質(zhì)中miR-146a表達(dá)升高,miR-146a表達(dá)量的變化可能與POCD的發(fā)病機(jī)制相關(guān)。
[Abstract]:Objective to observe the changes of miR-146a expression in serum, hippocampus and prefrontal cortex of postoperative cognitive dysfunctional POCD mice after operation. The body weight was 2530g. It was randomly divided into two groups: control group (C group) and anaesthesia operation group (as group). 25 rats in each group were treated with open reduction and internal fixation of tibial fracture under 2.1% isoflurane inhalation anesthesia. After 24 hours of operation, 3 mice in each group were taken for eyeball extraction, and blood was taken from each group 24 hours after operation. The expression of miR-146a in serum, hippocampus and prefrontal cortex of mice was measured by RT-PCR method. Ten mice in each group were collected on the 1st day after operation. Results compared with group C, the percentage of stiffness time in as group was significantly lower than that in group C (P 0.05). The results showed that there was no significant difference in percentage of stiffness time between the two groups in fear memory experiment. Compared with group C, the expression of miR-146a in as group was significantly higher than that in group C, and the expression of miR-146a in group as was significantly lower than that in group C at 12 h after operation. In hippocampus and group C, the expression of miR-146a in group as was significantly higher than that in group C, and the expression of miR-146a in group as was significantly higher than that in group C at 12 h after operation, and was significantly higher than that in group C at 12 h after operation, and at 12 h after operation, the expression of miR-146a in as group was significantly lower than that in group C at 12 h after operation, and compared with that in group C at 12 h after operation. In the prefrontal cortex, the expression of miR-146a in the prefrontal cortex was significantly increased at 24 hours after operation in as group, and the expression of miR-146a in as group was significantly higher than that in 24 h after operation at 24 h after operation. Conclusion the expression of miR-146a in as group was significantly higher than that in control group at 24 h after operation. Conclusion the expression of miR-146a in the serum of POCD mice is significantly higher than that in the control group. The increase of miR-146a expression in hippocampus and prefrontal cortex may be related to the pathogenesis of POCD.
