【摘要】：目的:檢測健康人群和骨關(guān)節(jié)結(jié)核患者外周血全血細(xì)胞Toll樣受體4(Toll-like receptor 4,TLR4)在mRNA水平上的相對表達(dá)量;同時檢測兩組受試者血清中腫瘤壞死因子-α(Tumor necrosis factor-α,TNF-α)和白細(xì)胞介素-6(Interleukin-6,IL-6)的表達(dá)情況,以初步探討骨關(guān)節(jié)結(jié)核外周血全血細(xì)胞TLR4表達(dá)情況和意義,以及其與血清TNF-α、IL-6水平的相關(guān)性。方法:(1)采用實時熒光定量PCR(Real Time PCR)技術(shù)檢測TLR4 m RNA在113名健康者和81例骨關(guān)節(jié)結(jié)核患者全血細(xì)胞中的相對表達(dá)量,比較兩組間的差異。(2)應(yīng)用雙抗體夾心免疫酶聯(lián)吸附法(Enzyme linked immunosorbent assay,ELISA)對兩組受試者血清中TNF-α和IL-6的表達(dá)含量進行檢測,比較兩組間的表達(dá)差異。(3)分析觀察組中TLR4 mRNA相對表達(dá)量與血清TNF-α、IL-6水平之間的相關(guān)性,以及血清TNF-α水平與IL-6水平的相關(guān)性。結(jié)果:(1)健康對照組TLR4 mRNA的相對表達(dá)量為0.417±0.082,骨結(jié)核結(jié)核患者TLR4 mRNA的相對表達(dá)量為0.735±0.161。兩組TLR4 mRNA相對表達(dá)量之間的差異具有統(tǒng)計學(xué)意義(P0.01)。(2)觀察組患者血清TNF-α和IL-6的表達(dá)水平均高于對照組,差異均具有統(tǒng)計學(xué)意義(P0.01)。(3)在觀察組中,TLR4 mRNA相對表達(dá)量與血清TNF-α、IL-6水平均無顯著線性相關(guān)(相關(guān)系數(shù)r=0.178,P0.05;相關(guān)系數(shù)r=0.043,P0.05);血清TNF-α水平與IL-6水平亦無明顯線性相關(guān)(相關(guān)系數(shù)r=-0.042,P0.05)。結(jié)論:(1)骨關(guān)節(jié)結(jié)核患者外周血全血細(xì)胞TLR4 mRNA相對表達(dá)量顯著高于健康人群,提示TLR4基因在mRNA水平上的高表達(dá)可能與骨關(guān)節(jié)結(jié)核的免疫調(diào)節(jié)存在密切聯(lián)系。(2)骨關(guān)節(jié)結(jié)核患者血清TNF-α和IL-6水平明顯高于健康人群,說明其可能在骨關(guān)節(jié)結(jié)核的發(fā)生發(fā)展中起著重要的作用。(3)在骨關(guān)節(jié)結(jié)核患者中,外周血全血細(xì)胞TLR4 mRNA相對表達(dá)量與血清TNF-α、IL-6水平均無顯著線性相關(guān)。
[Abstract]:Objective: to detect the relative expression of Toll like receptor 4Toll-like receptor 4 (TLR4) in peripheral blood of healthy people and patients with bone and joint tuberculosis at the level of mRNA, and to detect the expression of tumor necrosis factor- 偽 Tumor necrosis factor- 偽 (TNF- 偽) and interleukin-6 Interleukin-6IL-6 (IL-6) in the serum of both groups. To explore the expression and significance of TLR4 in peripheral blood of osteoarticular tuberculosis. Methods the relative expression of TLR4 m RNA in the whole blood cells of 113 healthy subjects and 81 patients with osteoarticular tuberculosis was detected by real-time fluorescence quantitative PCR(Real Time PCR. The expression of TNF- 偽 and IL-6 in serum of the two groups was detected by double antibody sandwich immunosorbent assay (Elisa). To analyze the correlation between the relative expression of TLR4 mRNA and the serum TNF- 偽 IL-6 level in the observation group. Results the relative expression of TLR4 mRNA in healthy control group was 0.417 鹵0.082, and the relative expression of TLR4 mRNA in tuberculosis of bone was 0.735 鹵0.161. The difference between the two groups was statistically significant. The serum levels of TNF- 偽 and IL-6 in the observation group were higher than those in the control group. There was no significant linear correlation between the relative expression of TLR4 mRNA and serum TNF- 偽 IL-6 level in the observation group (r = 0.178, P 0.05, r = 0.043, P 0.05, r = 0.043, P 0.05, respectively), and there was no significant linear correlation between the serum TNF- 偽 level and the level of IL-6 (correlation coefficient). Conclusion the relative expression of TLR4 mRNA in peripheral blood of patients with osteoarticular tuberculosis was significantly higher than that of healthy controls. It is suggested that the high expression of TLR4 gene at mRNA level may be closely related to the immunomodulation of osteoarticular tuberculosis.) Serum TNF- 偽 and IL-6 levels in patients with osteoarticular tuberculosis are significantly higher than those in healthy controls. It is suggested that the relative expression of TLR4 mRNA in peripheral blood cells is not linearly correlated with the serum TNF- 偽 IL-6 level in patients with osteoarticular tuberculosis, which may play an important role in the occurrence and development of osteoarticular tuberculosis.
