【摘要】：目的探討高濃度褪黑素抑制人骨肉瘤細(xì)胞系MG-63細(xì)胞增殖的潛在機制。方法在體外培養(yǎng)的MG-63細(xì)胞中,外源性加入4 mmol/L濃度褪黑素,分別用Western blotting和實時PCR方法檢測高濃度褪黑素對細(xì)胞周期蛋白(cyclins)及其依賴性激酶(CDK)蛋白和m RNA水平表達的影響。結(jié)果 Western blotting和實時PCR檢測結(jié)果顯示,4 mmol/L濃度的褪黑素以時間依賴性方式顯著地下調(diào)了與細(xì)胞周期G_1期進展有關(guān)的cyclin D1和CDK4蛋白和m RNA表達水平,與細(xì)胞周期G_2/M期進展有關(guān)的cyclin B1和CDK1蛋白和m RNA表達水平,但與G_1/S期轉(zhuǎn)變和S期進展有關(guān)的cyclin E、CDK2和cyclin A蛋白和m RNA表達水平?jīng)]有變化。結(jié)論褪黑素以時間依賴性方式,通過誘導(dǎo)細(xì)胞周期停滯抑制骨肉瘤MG-63細(xì)胞增殖,這種抑制效應(yīng)與cyclin D1和CDK4及cyclin B1和CDK1的表達下調(diào)有關(guān)。
[Abstract]:Objective to investigate the potential mechanism of high concentration of melatonin in inhibiting proliferation of human osteosarcoma cell line MG-63. Methods in vitro cultured MG-63 cells were treated with 4 mmol/L melatonin. Western blotting and real time PCR were used to detect the effects of melatonin on the expression of cyclin cyclins and their dependent kinases, and the expression of m RNA. Results Western blotting and real time PCR assays showed that melatonin 4 mmol/L concentration was melatonin. The expression levels of cyclin D1 and CDK4 proteins and m RNA related to the progression of cell cycle G1 were significantly down-regulated in a time-dependent manner. The expression levels of cyclin B1 and CDK1 protein and m RNA related to the progression of G2 / M phase in cell cycle were not changed, but the expression levels of cyclin ECDK2 and cyclin A protein and m RNA related to G1- S phase transition and S phase progression were not changed. Conclusion melatonin is time-dependent. The proliferation of osteosarcoma MG-63 cells was inhibited by cell cycle arrest, which was related to the down-regulation of the expression of cyclin D1 and CDK4 and cyclin B1 and CDK1.
