調(diào)節(jié)軟骨細(xì)胞骨架蛋白對成軟骨表型的影響

【摘要】:目的研究軟骨細(xì)胞骨架蛋白結(jié)構(gòu)改變與成軟骨細(xì)胞表型改變的關(guān)系。方法采用三維微團(tuán)培養(yǎng)法培養(yǎng)新西蘭兔膝關(guān)節(jié)軟骨細(xì)胞,將軟骨細(xì)胞分為對照組、秋水仙堿組、細(xì)胞松弛素B組及丙烯酰胺組,通過秋水仙堿、細(xì)胞松弛素B及丙烯酰胺分別調(diào)節(jié)軟骨細(xì)胞骨架的微管、肌動蛋白微絲及波形蛋白中間纖絲,從大體形態(tài)、組織切片染色、免疫組化染色及RT-PCR分析成軟骨細(xì)胞表型的改變。結(jié)果對照組、秋水仙堿組及細(xì)胞松弛素B組細(xì)胞較密集、均一,丙烯酰胺組細(xì)胞較少、分散。對照組、秋水仙堿組及細(xì)胞松弛素B組COLⅡ、蛋白多糖染色深,COLⅠ染色淺;而丙烯酰胺組COLⅡ、蛋白多糖染色淺,COLⅠ染色較深。RT-PCR數(shù)據(jù)統(tǒng)計分析顯示對照組與秋水仙堿組、細(xì)胞松弛素B組相比,COLⅠ(1.21±0.21 vs 1.55±0.06、1.70±0.07)及COLⅡ(1.10±0.12 vs 0.91±0.21、0.90±0.30)表達(dá)差異無統(tǒng)計學(xué)意義(P0.05);而對照組COLⅡ表達(dá)明顯多于丙烯酰胺組(0.26±0.06),COLⅠ表達(dá)明顯低于丙烯酰胺組(9.63±0.38),差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論丙烯酰胺調(diào)節(jié)軟骨細(xì)胞骨架波形蛋白中間纖絲可明顯下調(diào)COLⅡ、蛋白多糖的表達(dá)。
[Abstract]:Objective to study the relationship between the structural changes of chondrocytoskeleton protein and the phenotypic changes of chondroblasts. Methods the chondrocytes of knee joint of New Zealand rabbits were cultured by three-dimensional microgroup culture and the chondrocytes were divided into control group. In colchicine group, cytochalasin B group and acrylamide group, the microtubules, actin microfilaments and vimentin intermediate filaments of chondrocyte cytoskeleton were regulated by colchicine, cytochalasin B and acrylamide, respectively. The changes of chondroblast phenotype were analyzed from gross morphology, tissue section staining, immunohistochemical staining and RT-PCR.
