【摘要】：目的制備非協(xié)調(diào)性分子5同源蛋白B(UNC5B)的單克隆抗體(mAb),分析其功能。方法將UNC5B基因片段與原核表達(dá)載體pET-32a連接,連接產(chǎn)物轉(zhuǎn)化大腸桿菌BL21(DE3),進(jìn)行原核誘導(dǎo)表達(dá)并純化重組蛋白。用UNC5B重組蛋白免疫BALB/c小鼠,經(jīng)雜交瘤技術(shù)篩選可穩(wěn)定分泌抗UNC5B的雜交瘤細(xì)胞株,并用Western blot法、ELISA和流式細(xì)胞術(shù)鑒定。利用細(xì)胞劃痕實驗觀察UNC5BmAb對黑素瘤細(xì)胞遷移能力的影響。結(jié)果表達(dá)并純化了UNC5B重組蛋白;篩選出1株高效價的2C9mAb,通過ELISA、Western blot法和流式細(xì)胞術(shù)證實該m Ab特異性識別UNC5B;劃痕實驗證明在netrin-1存在的條件下,UNC5BmAb可促進(jìn)黑素瘤細(xì)胞遷移。結(jié)論成功制備了UNC5B的特異性m Ab,在netrin-1存在時,該抗體可促進(jìn)黑素瘤細(xì)胞遷移。
[Abstract]:Objective to prepare monoclonal antibody (mAb), to analyze the function of discordant molecule 5 homologous protein B (UNC5B). Methods the UNC5B gene fragment was ligated with prokaryotic expression vector pET-32a, and the ligated product was transformed into E. coli BL21 (DE3). The recombinant protein was induced and purified. BALB/c mice were immunized with UNC5B recombinant protein. Hybridoma cell lines stably secreting anti-UNC5B were screened by hybridoma technique and identified by Western blot assay, ELISA and flow cytometry. The effect of UNC5BmAb on the migration ability of melanoma cells was observed by cell scratch test. Results the recombinant protein of UNC5B was expressed and purified, and a high valence 2C9mAbstrain was screened out. The specific recognition of UNC5B; by mAb was confirmed by ELISA,Western blot and flow cytometry. Scratch test showed that UNC5BmAb could promote the migration of melanoma cells in the presence of netrin-1. Conclusion the specific m Ab, of UNC5B can promote the migration of melanoma cells in the presence of netrin-1.
