【摘要】：目的探討咪喹莫特誘導(dǎo)的銀屑病鼠(IIPM)模型皮損角質(zhì)形成細(xì)胞(KCs)STAT3的表達(dá)和活化STAT3(p-STAT3)的水平及其意義。方法采用免疫組織化學(xué)Polymer酶標(biāo)二抗法,檢測(cè)16例IIPM皮損和16例取材于對(duì)照鼠正常皮膚組織KCs中STAT3的表達(dá)和p-STAT3的水平,并利用PASI對(duì)IIPM皮損進(jìn)行臨床積分(TCS)評(píng)價(jià)。結(jié)果 IIPM皮損KCs中STAT3表達(dá)顯著高于對(duì)照鼠正常皮膚組織(后者呈低表達(dá))(P=0.018),且其p-STAT3水平亦顯著高于后者(后者呈低水平)(P=0.003);IIPM皮損KCs STAT3表達(dá)、p-STAT3水平均與IIPM皮損TCS之間存在正相關(guān)(rs=0.58,0.76;P=0.019,0.001),隨著STAT3表達(dá)的增加、p-STAT3水平的升高,皮損TCS也呈升高趨勢(shì)。結(jié)論 KCs STAT3的表達(dá)增加和活化對(duì)IIPM模型銀屑病樣皮膚炎癥性皮損的形成可能起重要作用,可利用IIPM模型進(jìn)行關(guān)于STAT3的研究。
[Abstract]:Objective to investigate the expression of KCsMS-STAT3 and the level of activated STAT3p-STAT3 in IIPM model of psoriatic rats induced by IIPM. Methods the expression of STAT3 and the level of p-STAT3 in KCs of 16 cases of IIPM skin lesions and 16 cases of normal skin tissues were detected by immunohistochemical Polymer enzyme-labeled antibody method, and the clinical integral TCSs were evaluated by PASI in IIPM skin lesions. Results the expression of STAT3 in KCs of IIPM lesions was significantly higher than that in normal skin tissues of control rats (the latter showed a low expression level of P0. 018, and the level of p-STAT3 was also significantly higher than that of the latter. The latter presented a low level of expression of KCs STAT3 in the lesions of IIPM and the expression of p-STAT3 in the lesions of IIPM and TCS of IIPM lesions. In the positive correlation, the level of p-STAT3 was increased with the increase of STAT3 expression, and the expression of STAT3 was increased with the increase of the expression of STAT3, and the level of p-STAT3 was increased with the increase of the expression of STAT3. TCS of lesions also showed an increasing trend. Conclusion the increased expression and activation of KCs STAT3 may play an important role in the formation of psoriatic inflammatory skin lesions in IIPM model.
