間歇低氧大鼠肝細(xì)胞GLUT-2、GCK表達(dá)變化與胰島素抵抗的相關(guān)性研究

【摘要】:目的:制備間歇低氧大鼠模型,檢測(cè)肝細(xì)胞葡萄糖轉(zhuǎn)運(yùn)蛋白-2(GLUT-2)、葡萄糖激酶(GCK)蛋白表達(dá)量變化及測(cè)定胰島素抵抗的程度,探討間歇低氧條件下GLUT-2、GCK的表達(dá)變化與機(jī)體胰島素抵抗發(fā)生的相關(guān)機(jī)制。方法:24只6周齡健康雄性SD大鼠隨機(jī)分為對(duì)照組(NC組)、間歇低氧4周組(IH4組)和間歇低氧8周組(IH8組),每組8只。造模組按預(yù)先設(shè)定的通氣模式給與間歇低氧暴露(9:00am-5:00pm),對(duì)照組給予間歇壓縮空氣,暴露時(shí)間同造模組。測(cè)定每組大鼠空腹血糖、空腹胰島素,穩(wěn)態(tài)模型評(píng)估-胰島素抵抗指數(shù)(HOMA-IR)及胰島素敏感指數(shù)(ISI)評(píng)估胰島素抵抗程度。免疫組織化學(xué)染色觀察肝細(xì)胞GLUT-2、GCK蛋白表達(dá)變化,測(cè)定平均灰度值對(duì)蛋白表達(dá)進(jìn)行定量分析,整合數(shù)據(jù)并統(tǒng)計(jì)分析。結(jié)果:1.與NC組相比,造模組空腹血糖、空腹胰島素、HOMA-IR升高,ISI降低,且隨暴露時(shí)間的延長(zhǎng),上述變化更趨明顯,差異有統(tǒng)計(jì)學(xué)意義(P均0.05)。2.間歇低氧條件下肝細(xì)胞GLUT-2、GCK蛋白表達(dá)量均降低,與4周組相比8周組變化更顯著,差異有統(tǒng)計(jì)學(xué)意義(P均0.05)。3.Pearson相關(guān)分析顯示:GLUT-2、GCK平均灰度值均與HOMA-IR呈正相關(guān)(r=0.894,r=0.869,P均0.05),與ISI呈負(fù)相關(guān)(r=-0.886,r=-0.906,P均0.05)。結(jié)論:1.間歇低氧暴露組大鼠空腹血糖、空腹胰島素水平及HOMA-IR升高,ISI降低,機(jī)體呈現(xiàn)糖代謝紊亂及胰島素抵抗征象,且隨著暴露的時(shí)間延長(zhǎng)上述變化趨于明顯;2.間歇低氧條件下肝細(xì)胞GLUT-2、GCK蛋白表達(dá)下調(diào),且兩蛋白所測(cè)灰度值與HOMA-IR相關(guān);3.肝細(xì)胞GLUT-2、GCK蛋白參與間歇低氧條件下肝胰島素抵抗的發(fā)生。
[Abstract]:Objective: to establish a rat model of intermittent hypoxia and to detect the expression of glucose transporter (GLUT-2), glucokinase (GCK) and the degree of insulin resistance (IR) in hepatocytes.To investigate the relationship between the expression of GLUT-2 GCK and insulin resistance under intermittent hypoxia.Methods Twenty four male Sprague-Dawley rats aged 6 weeks were randomly divided into control group (NC group), intermittent hypoxia group (group IH4) and intermittent hypoxia group (group IH8, 8 rats in each group).The model group was given intermittent hypoxia exposure (9: 00am-5: 00pmg) according to the preset ventilation mode, while the control group was given intermittent compressed air. The exposure time was the same as the model group.Fasting blood glucose, fasting insulin, homeostasis model assessment-insulin resistance index (HOMA-IRI) and insulin sensitivity index (ISI) were measured in each group.The expression of GLUT-2 GCK in hepatocytes was observed by immunohistochemical staining. The expression of GLUT-2GCK was quantitatively analyzed by measuring the mean gray value, and the data were integrated and analyzed statistically.The result is 1: 1.Compared with NC group, fasting blood glucose, fasting insulin and HOMA-IR in model group increased and ISI decreased. With the prolongation of exposure time, the above changes were more obvious, and the difference was statistically significant (P < 0.05).Conclusion 1.In intermittent hypoxic exposure group, fasting blood glucose, fasting insulin level and HOMA-IR were increased and decreased, the body showed glucose metabolism disorder and insulin resistance signs, and the above changes tended to be obvious with the prolongation of exposure time.The expression of GLUT-2 GCK protein was down-regulated in hepatocytes under intermittent hypoxia, and the gray values of the two proteins were correlated with HOMA-IR.GLUT-2 GCK protein is involved in the development of insulin resistance in the liver under intermittent hypoxia.
