【摘要】：目的研究內(nèi)源性物質(zhì)3-硫酸吲哚酚用于腎損傷早期診斷的意義。方法以抗癌藥順鉑、甲氨蝶呤為研究對象,取雄性Sprague-Dawley大鼠36只,隨機分為3組,分別為順鉑組、甲氨蝶呤組和空白組。于給藥后第1、3和5天采集血清與新鮮腎組織。采用LC-MS法測定各組大鼠血清中3-硫酸吲哚酚的質(zhì)量濃度;同時分析各組大鼠血清中生化指標肌酐和尿素氮濃度以及腎臟組織病理學變化。結(jié)果單次給藥后,腎組織病理學結(jié)果顯示順鉑組與甲氨蝶呤組大鼠在第3天均可見明顯腎損傷;各組中內(nèi)源性物質(zhì)3-硫酸吲哚酚于第3天明顯升高;順鉑組肌酐與尿素氮于第5天明顯升高,甲氨蝶呤組肌酐與尿素氮無明顯變化。結(jié)論在順鉑和甲氨蝶呤致腎損傷模型中,內(nèi)源性物質(zhì)3-硫酸吲哚酚顯示了較常規(guī)生化指標血肌酐與尿素氮更高的靈敏度。
[Abstract]:Objective to study the value of endogenous substance 3-indophenol sulfate in early diagnosis of renal injury. Methods 36 male Sprague-Dawley rats were randomly divided into three groups: cisplatin group, methotrexate group and blank group. Serum and fresh renal tissue were collected on the 1st, 3rd and 5th day after administration. The concentration of indolephenol 3-sulfate in serum of rats in each group was determined by LC-MS and the biochemical indexes creatinine and urea nitrogen in serum and histopathological changes of kidney in each group were analyzed at the same time. Results after single administration, renal histopathology showed that both cisplatin group and methotrexate group had obvious renal injury on the 3rd day, the endogenous substance 3-indophenol sulfate significantly increased on the 3rd day, and the renal injury was observed on the 3rd day after the administration of cisplatin and methotrexate. Creatinine and urea nitrogen increased significantly in cisplatin group on the 5th day, but there was no significant change in methotrexate group. Conclusion in the model of renal injury induced by cisplatin and methotrexate, the endogenous substance 3-indophenol sulfate shows a higher sensitivity than routine biochemical markers such as serum creatinine and urea nitrogen.
