【摘要】：目的探討辣椒素對(duì)膀胱癌細(xì)胞系253J與RT112侵襲轉(zhuǎn)移的影響。方法以0、100、200μmol/L辣椒素分別作用于人膀胱癌253J與RT112細(xì)胞0、12、24 h,采用MTT法檢測(cè)253J與RT112細(xì)胞活力,劃痕實(shí)驗(yàn)檢測(cè)253J與RT112細(xì)胞遷移能力,Transwell法檢測(cè)253J與RT112細(xì)胞侵襲能力。Western blotting法檢測(cè)上皮間充質(zhì)轉(zhuǎn)化相關(guān)蛋白(E-鈣黏蛋白,N-鈣黏蛋白,波形蛋白)和Nrf2相關(guān)蛋白的表達(dá)水平。結(jié)果與0μmol/L比較,在100、200μmol/L辣椒素作用下,253J與RT112各組細(xì)胞活性降低(P0.01),侵襲能力下降(P0.01),遷移能力也下降(253J:P0.01;RT112:P=0.026或P=0.014)。細(xì)胞E-鈣黏蛋白、C-nrf2表達(dá)上升,而波形蛋白與N-nrf2蛋白表達(dá)減弱,并呈劑量-時(shí)間依賴性。結(jié)論辣椒素可抑制人膀胱癌細(xì)胞的侵襲與轉(zhuǎn)移,該作用可能與Nrf2的調(diào)節(jié)有關(guān)。
[Abstract]:Objective to investigate the effect of capsaicin on invasion and metastasis of bladder cancer cell line 253J and RT112. Methods Human bladder cancer cell line 253J and RT112 cells were treated with 0100200渭 mol / L capsaicin for 0,12,24 h, respectively. The viability of 253J and RT112 cells was detected by MTT assay, and the migration ability of 253J and RT112 cells was detected by scratch assay. The invasiveness of 253J and RT112 cells was detected by Transwell, and the expression of E-cadherin (N-cadherin, vimentin) and Nrf2-associated protein were detected by Western blotting. Results compared with 0 渭 mol / L capsaicin, the cell activity, invasion ability and migration ability of 253J and RT112 groups were decreased (P0.01), and the migration ability was also decreased (253J / mol / L capsaicin 0.026 or 0.014). The expression of E-cadherin and C-nrf2 increased, but the expression of vimentin and N-nrf2 decreased in a dose-time dependent manner. Conclusion capsaicin can inhibit the invasion and metastasis of human bladder cancer cells, which may be related to the regulation of Nrf2.
