【摘要】：目的：探討Galanin及Galanin2型受體(Ga1R2)在前列腺良惡性組織中的表達及其臨床意義。 方法：我們收集50例前列腺增生(BPH)、50例未轉(zhuǎn)移前列腺癌(PCa)及30例發(fā)生骨轉(zhuǎn)移前列腺癌(BM)標(biāo)本,運用免疫組織化學(xué)法觀察Galanin及Ga1R2在這些組織中的表達,并分析其臨床意義。我們選取人前列腺增生細胞株BPH-I、前列腺癌細胞株DU145為研究對象,兩組細胞培養(yǎng)后,提取細胞總蛋白,然后通過Western印跡法分別檢測兩組細胞株中Galanin及Ga1R2蛋白表達情況。 結(jié)果：免疫組化結(jié)果顯示,Galanin蛋白陽性表達在BPH組為9例,PCa組為34例,BM組為25例；GalR2蛋白陽性表達在BPH組為40例,PCa組為7例,BM組為2例。結(jié)果顯示Galanin在PCa組及BM組蛋白表達陽性率明顯高于BPH組,組間比較差異均有統(tǒng)計學(xué)意義(χ2值分別為25.50和32.75,均P0.01),Galanin在PCa組與BM組間蛋白表達陽性率無統(tǒng)計學(xué)差異(χ2＝2.28,P0.05)；而GalR2在BPH組蛋白表達陽性率明顯高于PCa組及BM組,組間比較差異均有統(tǒng)計學(xué)意義(χ2值分別為43.72和40.43,均P0.01),在PCa組與BM組問蛋白表達陽性率比較差異無統(tǒng)計學(xué)意義(χ2=1.01,P0.05)。在PCa組及BM組內(nèi),Galanin與Ga1R2的蛋白表達均呈顯著負相關(guān)(rs=-0.55,P0.01；rs=-0.77,P0.01)。Western印跡法結(jié)果顯示,Galanin蛋白在BPH-1細胞中的表達明顯減少,在DU145細胞中的表達增多；相反,GalR2蛋白在BPH-I細胞中的表達增多,在DU145細胞中的表達明顯減少。此結(jié)果與免疫組化結(jié)果一致。 結(jié)論：Galanin在前列腺癌組織中的蛋白陽性表達率高,提示Galanin可能參與了前列腺癌的發(fā)病機制；而GalR2在前列腺癌組織中蛋白表達水平降低,免疫組化結(jié)果中GalR2與Galanin蛋白表達水平呈負相關(guān),提示Galanin在前列腺癌中可能是通過與Ga1R2之外的兩種受體相結(jié)合從而發(fā)揮生物學(xué)作用,也提示Galanin與Ga1R2兩種蛋白水平的聯(lián)合檢測對前列腺良惡性病變的診斷具有較好的臨床意義。
[Abstract]:Objective: to investigate the expression and clinical significance of Galanin and Galanin2 receptor (Ga1R2) in benign and malignant prostate tissues. Methods: 50 cases of benign prostatic hyperplasia (BPH) and 50 cases of (PCa) without metastasis and 30 cases of (BM) with bone metastasis were collected. The expression of Galanin and Ga1R2 in these tissues was observed by immunohistochemical method and its clinical significance was analyzed. We selected human prostatic hyperplasia cell line BPH-I, prostate cancer cell line DU145 as the research object. After two groups of cells were cultured, the total cell protein was extracted, and then the expression of Galanin and Ga1R2 protein in the two groups were detected by Western blot. Results: immunohistochemical results showed that the positive expression of Galanin protein in BPH group (9 cases) was 34 cases in BM group (34 cases) and in BPH group (40 cases), 7 cases (7 cases) in BPH group, 2 cases in BM group. The results showed that the positive rate of Galanin expression in PCa group and BM group was significantly higher than that in BPH group (蠂 2 = 25.50 and 32.75, respectively, P0.01). There was no significant difference between PCa group and BM group (蠂 2 / 2.28 P 0.05). The positive rate of GalR2 expression in BPH group was significantly higher than that in PCa group and BM group (蠂 ~ 2 = 43.72 and 40.43, respectively, P0.01). There was no significant difference between PCa group and BM group (蠂 ~ (2) 1.01). In PCa group and BM group, the expression of Galanin and Ga1R2 protein was negatively correlated (rs=-0.55,P0.01;rs=-0.77,P0.01). Western blotting showed that the expression of Galanin protein in BPH-1 cells decreased significantly, but increased in DU145 cells, on the contrary, the expression of Galanin protein in BPH-I cells increased. The expression in DU145 cells was significantly decreased. The results were consistent with the immunohistochemical results. Conclusion the positive expression rate of Galanin in prostate cancer tissues is high, suggesting that Galanin may be involved in the pathogenesis of prostate cancer, while the protein expression level of GalR2 in prostate cancer tissue is decreased. The expression of GalR2 and Galanin protein was negatively correlated with immunohistochemical results, suggesting that Galanin may play a biological role in prostate cancer by combining with two receptors other than Ga1R2. It is also suggested that the combined detection of Galanin and Ga1R2 protein levels has a good clinical significance in the diagnosis of benign and malignant prostate diseases.
