【摘要】：目的研究泛素連接酶Parkin在糖尿病腎病(diabetic nephropathy,DN)患者腎組織中的表達規(guī)律及與腎小管間質(zhì)損傷的關系。方法納入2015-2016年我院收治的40例經(jīng)腎活檢確診為2型DN患者,并取10例腎腫瘤切除后遠端正常腎組織為對照組。采用免疫組化檢測不同DN間質(zhì)損傷評分腎組織的Parkin表達水平,并與臨床指標、腎組織病理損傷進行相關性分析。免疫熒光共染檢測腎小管Parkin的表達分別與腎間質(zhì)損傷因子IL-6、TGF-β的表達及間質(zhì)纖維化標志Col-Ⅳ的表達的關系。結果 Parkin主要在腎小管上皮細胞細胞質(zhì)中表達,并隨著DN腎小管間質(zhì)損傷評分的加重而逐漸減少。Parkin表達與腎臟結構損傷(腎小球硬化、腎小管萎縮與間質(zhì)纖維化、腎間質(zhì)炎癥)評分呈顯著負相關(P0.01),與腎臟功能損傷指標[尿蛋白定量、尿N-乙酰-β-氨基葡萄糖苷酶(NAG)、尿素氮、血清胱抑素C、血肌酐]呈負相關關系(P0.01),與腎小球濾過率呈正相關關系(P0.05)。腎組織Parkin陽性腎小管細胞并不表達促炎因子IL-6、促纖維化因子TGF-β及Col-Ⅳ。結論 Parkin表達降低與DN腎間質(zhì)損傷密切相關。
[Abstract]:Objective to study the expression of ubiquitin ligase Parkin in renal tissue of diabetic nephropathy patients and its relationship with tubulointerstitial injury. Methods Forty patients with type 2 DN diagnosed by renal biopsy were enrolled in our hospital from 2015 to 2016, and 10 cases of normal distal renal tissue after nephrectomy were taken as control group. Immunohistochemistry was used to detect the expression of Parkin in renal tissue with different DN interstitial injury scores, and the correlation with clinical indexes and pathological injury of renal tissue was analyzed.
