磷酸甘油酸變位酶1在生精功能障礙睪丸組織中的表達(dá)及意義

【摘要】:目的:檢測磷酸甘油酸變位酶1(PGAM1)在不同程度生精功能不良睪丸組織中的表達(dá)情況,并分析PGAM1表達(dá)與臨床病理參數(shù)的關(guān)系。方法:收集生精功能正常、輕度生精功能不良、重度生精功能不良、唯支持細(xì)胞綜合征患者睪丸組織標(biāo)本,統(tǒng)計性激素六項結(jié)果,采用免疫組化、qRT-PCR檢測PGAM1在不同程度生精功能不良睪丸組織中的表達(dá)情況。結(jié)果:免疫組化結(jié)果顯示PGAM1的高表達(dá)率在生精功能正常、輕度生精功能不良、重度生精功能不良及唯支持細(xì)胞綜合征患者各組間比較,差異具有統(tǒng)計學(xué)意義(P=0.000),PGAM1表達(dá)與患者的年齡、睪丸體積、LH及T表達(dá)均無明顯相關(guān)(P0.05);而PGAM1低表達(dá)患者FSH水平高于PGAM1高表達(dá)患者(P=0.002 3);qRT-PCR結(jié)果提示PGAM1 mRNA在生精功能正常及唯支持細(xì)胞綜合征患者睪丸組織中表達(dá)水平明顯高于重度生精不良睪丸組織中的表達(dá),差異具有統(tǒng)計學(xué)意義(P=0.000),PGAM1 mRNA表達(dá)與患者的年齡、睪丸體積、LH及T表達(dá)均無明顯相關(guān)(P0.05),而在FSH≤12 mIU/mL患者睪丸組織中的表達(dá)明顯高于FSH12 mIU/mL患者(P=0.043 0)。結(jié)論:PGAM1的表達(dá)與睪丸生精功能障礙有關(guān),其低表達(dá)與重度生精功能不良的形成相關(guān),其異常高表達(dá)與唯支持細(xì)胞綜合征的形成有關(guān)。
[Abstract]:Aim: to detect the expression of phosphoglyceric acid mutase 1 (PGAM1) in testis with different degrees of spermatogenic dysfunction and to analyze the relationship between the expression of PGAM1 and clinicopathologic parameters. Methods: normal spermatogenic function, mild spermatogenic dysfunction and severe spermatogenic dysfunction were collected from testicular tissues of patients with Sertoli cell syndrome. Immunohistochemical qRT-PCR was used to detect the expression of PGAM1 in testicular tissues with different degrees of spermatogenic dysfunction. Results: Immunohistochemical results showed that the high expression rate of PGAM1 was compared among the patients with normal spermatogenic function, mild spermatogenic dysfunction, severe spermatogenic dysfunction and Sertoli cell syndrome. The difference was statistically significant between the expression of PGAM1 and the age of the patients. There was no significant correlation between the expression of LH and T in testicular volume, while the FSH level in patients with low expression of PGAM1 was higher than that in patients with high expression of PGAM1. The results of RT-PCR showed that the expression of PGAM1 mRNA in testicular tissues of patients with normal spermatogenic function and Sertoli's syndrome was clear.
