基于蛋白質(zhì)組學(xué)的尿液中腎臟疾病相關(guān)蛋白的篩選

【摘要】:目的:通過(guò)對(duì)正常人血漿、尿液、腎組織及腎臟疾病患者尿液蛋白質(zhì)組學(xué)獲取的數(shù)據(jù)進(jìn)行比較分析,尋找尿液中與腎臟疾病及腎臟損傷相關(guān)的生物標(biāo)志物。方法:腎臟疾病患者尿液蛋白質(zhì)組數(shù)據(jù)來(lái)自表現(xiàn)為腎病綜合征的膜性腎病(MN)和局灶節(jié)段性腎小球硬化(FSGS)患者尿液蛋白質(zhì)組譜圖,從Peptide Atlas 2013數(shù)據(jù)庫(kù)獲得正常人血漿、尿液和腎組織蛋白質(zhì)組數(shù)據(jù)。尋找腎臟疾病患者尿液蛋白質(zhì)組相對(duì)正常人尿液蛋白質(zhì)組富集蛋白,將篩選到的蛋白定義為腎臟疾病相關(guān)蛋白。尋找正常人血漿和尿液蛋白質(zhì)組中不出現(xiàn),同時(shí)在腎臟疾病患者尿液和腎組織蛋白質(zhì)組中出現(xiàn),且相對(duì)正常人尿液蛋白質(zhì)組富集的蛋白,將篩選到的蛋白定義為腎損傷相關(guān)蛋白。結(jié)果:篩選到40種腎臟疾病相關(guān)蛋白。發(fā)現(xiàn)4種與腎損傷相關(guān)的蛋白,分別為泛素-60s核糖體蛋白L40、S相激酶相關(guān)蛋白1、轉(zhuǎn)化生長(zhǎng)因子βⅡ型受體、谷胱甘肽S轉(zhuǎn)移酶A3。結(jié)論:通過(guò)對(duì)腎臟疾病患者尿液蛋白質(zhì)組學(xué)數(shù)據(jù)與正常人血漿、尿液和腎組織的蛋白質(zhì)組學(xué)數(shù)據(jù)進(jìn)行比較分析,從患者尿液中獲得了40種與腎臟疾病相關(guān)的蛋白和4種腎臟組織損傷相關(guān)的蛋白。上述蛋白成分在腎臟疾病診斷及病情判斷中的意義有待進(jìn)一步研究闡明。
[Abstract]:Objective: to find the biomarkers related to renal disease and renal injury by comparing and analyzing the proteomics data of normal human plasma, urine, renal tissue and renal diseases. Methods: the urine proteome data of patients with renal disease were obtained from the Peptide Atlas 2013 database of patients with membranous nephropathy (MNs) and focal segmental glomerulosclerosis (FSGS). Proteome data of urine and renal tissue. In order to find the proteome protein rich protein in urine of patients with renal disease, the selected protein is defined as the kidney disease related protein. It is found that there is no proteome in normal human plasma and urine, and it is also found in proteome of urine and renal tissue in patients with renal disease, and it is relatively rich in proteome in urine of normal people. The selected proteins are defined as kidney injury related proteins. Results: 40 kinds of renal disease related proteins were screened.
