【摘要】：目的:觀察腎毒性物質(zhì)硫酸吲哚酚對(duì)人內(nèi)皮祖細(xì)胞(EPC)增殖的影響。方法:體外培養(yǎng)EPC,分別用0(對(duì)照)、25、50、100和200 mg/L硫酸吲哚酚刺激24、72、120 h后,MTT法測定細(xì)胞活性;分別用0(對(duì)照)、200 mg/L硫酸吲哚酚刺激24 h后,Brd U法測定細(xì)胞增殖率,流式細(xì)胞術(shù)測定活性氧(ROS)。結(jié)果:硫酸吲哚酚體外可呈劑量和時(shí)間依賴性地抑制EPC細(xì)胞活性。200 mg/L硫酸吲哚酚作用24 h,EPC增殖受抑(P0.05),同時(shí)細(xì)胞內(nèi)ROS大量累積(P0.05)。結(jié)論:硫酸吲哚酚可通過增加EPC細(xì)胞內(nèi)ROS累積,抑制EPC增殖。
[Abstract]:Objective: to observe the effect of indole sulfate, a nephrotoxic substance, on the proliferation of human endothelial progenitor cells (EPCs). Methods: EPCs were cultured in vitro. The cell proliferation rate was measured by BrdU method after 24 h stimulation with 0 (control group) 200 mg/L indoleol sulfate. Results: Indole sulfate inhibited the proliferation of EPC cells in a dose-and time-dependent manner in a dose-and time-dependent manner. The proliferation of EPC cells was inhibited by indole sulfate for 24 h, and a large amount of ROS accumulated in the cells. Conclusion: Indole sulfate can inhibit the proliferation of EPC cells in a dose-and time-dependent manner.
