利用顯微技術(shù)對體外誘導(dǎo)巨噬細(xì)胞與腎結(jié)石晶體相互作用的形態(tài)學(xué)研究

【摘要】:目的:研究體外細(xì)胞培養(yǎng)體系中人巨噬細(xì)胞U937與腎結(jié)石晶體的相互作用,觀察細(xì)胞與晶體反應(yīng)的形態(tài)學(xué)改變,并探討巨噬細(xì)胞在腎結(jié)石形成中的可能作用。 方法:體外利用PMA(Phorbol-12-myristate-13-acetate,,PMA)對人單核細(xì)胞株U937進(jìn)行誘導(dǎo),激活分化為巨噬細(xì)胞后,并使用不同種類的腎結(jié)石晶體與之進(jìn)行共培養(yǎng)。分別運(yùn)用倒置相差顯微鏡、掃描電子顯微鏡及活細(xì)胞成像技術(shù)—顯微延時攝影靜態(tài)及動態(tài)地觀察對細(xì)胞與晶體的相互作用。根據(jù)細(xì)胞與不同種類晶體共培養(yǎng)的時段不同分組,采用茜素紅S—氯化十六烷基吡啶鈣鹽半定量測定法測定晶體-細(xì)胞反應(yīng)后細(xì)胞基質(zhì)的鈣鹽沉積程度。 結(jié)果:光鏡、掃描電鏡下可見巨噬細(xì)胞吸附、吞噬晶體。活細(xì)胞成像技術(shù)—顯微延時攝影捕捉到細(xì)胞吸附、吞噬、溶解及轉(zhuǎn)運(yùn)晶體的動態(tài)過程。茜素紅S—氯化十六烷基吡啶鈣鹽半定量測定法:羥基磷灰石(hydroxyapatite,HAP)與一水草酸鈣(calcium oxalate monohydrate,COM)混合實(shí)驗(yàn)組與其它組在相同時段細(xì)胞基質(zhì)的鈣鹽沉積程度差別有統(tǒng)計學(xué)意義(P<0.05),24小時組的程度最高;HAP組與空白對照組在相同時段差別有統(tǒng)計學(xué)意義(P<0.05),與COM組差別無統(tǒng)計學(xué)意義(P>0.05)?瞻讓φ战M與COM組在相同時段差別無統(tǒng)計學(xué)意義(P>0.05);各晶體實(shí)驗(yàn)組12小時組與24小時組后差別有統(tǒng)計學(xué)意義(P<0.05),6小時組分別與另外兩個時段組差別無統(tǒng)計學(xué)意義(P>0.05);不同實(shí)驗(yàn)組不同時段不存在交互作用(P>0.05)。結(jié)論:巨噬細(xì)胞具有粘附、吞噬及轉(zhuǎn)運(yùn)腎結(jié)石晶體的能力。HAP可能存在介導(dǎo)巨噬細(xì)胞增強(qiáng)吞噬HAP和COM的作用;巨噬細(xì)胞可能通過吞噬HAP后啟動吞噬COM。
[Abstract]:Aim: to study the interaction between human macrophage U937 and renal stone crystals in vitro, observe the morphological changes of cell and crystal reaction, and explore the possible role of macrophages in the formation of renal calculi. Methods: human mononuclear cell line U937 was induced by PMA-Phorbol-12-Eritrestate-13-acetatePMA in vitro, then activated and differentiated into macrophages and co-cultured with different kinds of kidney stone crystals. Scanning electron microscopy and living cell imaging techniques-static and dynamic observation of the interaction between cells and crystals by time-delay photography. The calcium deposition degree of cell matrix after crystal-cell reaction was determined by alizarin red S-cetylpyridine calcium salt semi-quantitative method. Results: under light microscope, macrophage adsorption and phagocytosis were observed under scanning electron microscope. Dynamic process of dissolution and transport of crystals. Semi-quantitative determination of alizarin red S- cetylpyridine chloride calcium salt: hydroxyapatite HAP) and calcium oxalate monohydrate monohydrate monohydrate. There was significant difference in sedimentation degree between HAP group and blank control group at the same time period (P < 0.05), but there was no significant difference between HAP group and COM group (P > 0.05). The difference between blank control group and COM group was at the same time. There was no statistical significance (P > 0.05), there was no significant difference between the experimental group and the other two groups after 12-hour group and 24-hour group (P < 0.05), there was no significant difference between the other two groups (P > 0.05), and there was no interaction between the experimental group and the other two groups (P > 0.05). Conclusion: macrophages have adhesion. The ability of phagocytosis and transport of renal stone crystals. HAP may mediate macrophages to enhance phagocytosis of HAP and COM, and macrophages may initiate phagocytosis of HAP and COM by phagocytosis of HAP.
