【摘要】：目的：采用免疫組織化學(xué)方法檢測(cè)SFRP2和β-catenin在前列腺增生和前列腺癌組織中的表達(dá)，以及腫瘤各臨床特征之間的關(guān)系。 方法：選自福建醫(yī)科大學(xué)附屬協(xié)和醫(yī)院前列腺增生及前列腺癌患者病理標(biāo)本各45例。采用免疫組化sp法檢測(cè)SFRP2蛋白和β-catenin的表達(dá)。采用SPSS13.0統(tǒng)計(jì)學(xué)軟件進(jìn)行分析SFRP2、β-catenin在腫瘤及增生組織中的表達(dá)差異及其與腫瘤各臨床特征之間的關(guān)系。 結(jié)果：在45例前列腺癌和45例前列腺增生組織中β-catenin蛋白的陽(yáng)性率分別為71.1%(32/45)和35.6%(16/45),卡方檢驗(yàn)組間差異有統(tǒng)計(jì)學(xué)意義（P0.01）；SFRP2蛋白陽(yáng)性率分別為24.4%（11/45）和66.7%(30/45),卡方檢驗(yàn)組間差異明顯（P0.01）。對(duì)45例前列腺癌患者術(shù)后病理分級(jí)、分期及年齡分組后分析β-catenin, SFRP2的表達(dá)關(guān)系示：年齡≥70歲患者β-catenin蛋白陽(yáng)性率61.2%，年齡＜70歲患者β-catenin蛋白陽(yáng)性率70%，，卡方檢驗(yàn)組間差異無(wú)統(tǒng)計(jì)學(xué)意義（P0.05）。同上分析SFRP2蛋白發(fā)現(xiàn)：SFRP2蛋白的陽(yáng)性表達(dá)與患者年齡之間差異得出p0.05，無(wú)統(tǒng)計(jì)學(xué)意義；前列腺癌中低分化組β-catenin蛋白的陽(yáng)性表達(dá)率明顯高于高分化組，低中分化組的SFRP2蛋白陽(yáng)性表達(dá)率明顯低于高分化組；在TNM分期中，前列腺癌的III+IV期分組β-catenin蛋白陽(yáng)性表達(dá)率高于I+II期分組，前列腺癌的III+IV期分組SFRP2蛋白的陽(yáng)性表達(dá)率則低于I+II期分組。Spearman等級(jí)相關(guān)分析發(fā)現(xiàn)β-catenin細(xì)胞膜表達(dá)缺失、異位表達(dá)均與SFRP2表達(dá)呈負(fù)相關(guān)(r=㧟0.474，P＜0.01；r=㧟0.565P＜0.01)；β-catenin細(xì)胞膜表達(dá)缺失與β-catenin異位表達(dá)呈正相關(guān)(r=0.741，P＜0.01)。 結(jié)論：1.前列腺增生組織β-catenin表達(dá)的陽(yáng)性率低于癌腫組織,SFRP2的陽(yáng)性表達(dá)高于癌腫組織，SFRP2和β-catenin蛋白的表達(dá)和前列腺癌的發(fā)生、發(fā)展密切相關(guān)。 2.前列腺增生組織中，β-catenin表達(dá)于前列腺細(xì)胞的細(xì)胞膜上,而SFRP2表達(dá)于前列腺細(xì)胞的細(xì)胞質(zhì)；在前列腺癌組織中，β-catenin則出現(xiàn)異位表達(dá)或者膜表達(dá)缺失，同時(shí)SFRP2表達(dá)則下調(diào)或減弱；β-catenin的異位表達(dá)可能是前列腺癌的早期事件，可成為前列腺癌的預(yù)防、早期診斷的標(biāo)記物。 3. SFRP2和β-catenin蛋白在前列腺癌組織中的表達(dá)與其病理分級(jí)、分期之間有明顯相關(guān)性,可進(jìn)一步評(píng)估患者預(yù)后。
[Abstract]:Aim: to detect the expression of SFRP2 and 尾 -catenin in prostatic hyperplasia and prostate cancer and the relationship between the clinical features of the tumor and the expression of 尾 -catenin by immunohistochemical method. Methods: the expressions of SFRP2 protein and 尾 -catenin were detected by immunohistochemical sp method in 45 cases of benign prostatic hyperplasia and 45 cases of prostate cancer in Union Hospital of Fujian Medical University. SFRP2and 尾 -catenin were analyzed by SPSS13.0 software. The difference of expression in tumor and proliferative tissue and its relationship with clinical features of tumor. Results: the positive rates of 尾 -catenin protein in 45 cases of prostate cancer and 45 cases of prostatic hyperplasia were 71.11 / 45 and 35.665 / 45, respectively. The positive rates of SFRP2 protein in the chi-square test group were 24.4 / 11 / 45 and 66.730 / 45, respectively.
