【摘要】：目的：目前臨床上對前列腺癌尤其進(jìn)展到激素非依賴性前列腺癌尚無有效治療方法,而藥物抗腫瘤尤其是天然植物在國內(nèi)外已成為腫瘤治療研究的熱點(diǎn)。近年來很多國內(nèi)外研究發(fā)現(xiàn)槲皮素這種天然植物提取物對很多腫瘤細(xì)胞均有抑制生長和抑制侵襲作用。本實(shí)驗(yàn)旨在通過槲皮素作用于激素非依賴性前列腺癌PC-3荷瘤鼠模型,觀察槲皮素對人前列腺癌PC-3裸鼠異種移植瘤的生長抑制作用效果,在此基礎(chǔ)上,探討槲皮素治療人前列腺癌PC-3細(xì)胞的作用和機(jī)制,為槲皮素治療激素非依賴性前列腺癌提供理論和實(shí)驗(yàn)依據(jù)。 方法:理論研究部分首先敘述中醫(yī)對前列腺癌的研究進(jìn)展,其次了解中西醫(yī)治療激素非依賴性前列腺癌的現(xiàn)狀,最后對槲皮素抗腫瘤作用機(jī)制作一綜述。實(shí)驗(yàn)研究部分首先體外培養(yǎng)人前列腺癌PC-3細(xì)胞,再制備小鼠激素非依賴性前列腺癌模型,槲皮素分組干預(yù)荷瘤鼠,最后測定不同組別小鼠移植瘤體積及重量；了解槲皮素對人前列腺癌PC-3裸鼠異種移植瘤的生長抑制作用效果,探討槲皮素治療激素非依賴性前列腺癌的具體機(jī)制。 結(jié)果：體內(nèi)裸鼠實(shí)驗(yàn)表明,槲皮素對激素非依賴性前列腺癌具有實(shí)驗(yàn)治療作用,并具有劑量依賴性,槲皮素低、中、高劑量組對人前列腺癌PC-3裸鼠移植瘤的相對腫瘤增殖率(T/C)分別為90.96%,72.46%,68.71%；抑瘤率達(dá)8.32%,24.80%,30.60%。 結(jié)論：槲皮素對人前列腺癌PC-3細(xì)胞裸鼠移植瘤的生長具有較顯著的抑制作用(P0.01),并具有劑量依賴性,表明槲皮素對激素非依賴性前列腺癌具有實(shí)驗(yàn)治療作用。
[Abstract]:Objective: at present, there is no effective treatment for prostate cancer, especially for hormone independent prostate cancer. In recent years, many researches at home and abroad have found that quercetin, a natural plant extract, has inhibited the growth and inhibition of many tumor cells. The aim of this study was to investigate the effects of quercetin on steroid-independent prostate cancer (PC-3) bearing mice. To observe the inhibitory effect of quercetin on the growth of xenograft tumor of human prostate cancer (PC-3) in nude mice, and to explore the effect and mechanism of quercetin on human prostate cancer PC-3 cells. To provide theoretical and experimental basis for quercetin in the treatment of hormone-independent prostate cancer. Methods: in the part of theoretical study, the research progress of traditional Chinese medicine (TCM) on prostate cancer and the current situation of hormone independent prostate cancer treated by traditional Chinese medicine (TCM) and western medicine (WM) were described. Finally, the anti-tumor mechanism of quercetin was reviewed. Firstly, human prostate cancer PC-3 cells were cultured in vitro, and then mouse steroid independent prostate cancer model was established. Quercetin was used to treat tumor bearing mice. The volume and weight of transplanted tumor were measured in different groups of mice. To investigate the effect of quercetin on the growth inhibition of xenograft tumor of human prostate cancer (PC-3) in nude mice, and to explore the mechanism of quercetin in the treatment of hormone independent prostate cancer. Results: in vivo experiments in nude mice showed that quercetin had an experimental therapeutic effect on hormone independent prostate cancer in a dose-dependent manner, and quercetin was low and moderate. The relative tumor proliferation rate (T / C) of the high dose group to transplanted human prostate cancer PC-3 nude mice was 90.96 / 72.46 / 68.71, respectively. The tumor inhibition rate was 8.32% and 24.80%, 30.60%. Conclusion: quercetin can inhibit the growth of human prostate cancer PC-3 cells in nude mice in a dose-dependent manner. It is suggested that quercetin has an experimental therapeutic effect on hormone-independent prostate cancer.
