白金養(yǎng)胃顆粒的藥效學(xué)研究

【摘要】:目的:1.白金養(yǎng)胃顆粒急性毒性實驗2、模擬大鼠幽門結(jié)扎型胃潰瘍模型,觀察白金養(yǎng)胃顆粒是否具有抗胃潰瘍作用,并研究白金養(yǎng)胃顆?刮笣冏饔脵C理:采用熱板法、小鼠耳腫脹法進行小鼠鎮(zhèn)痛和抗炎實驗,觀察白金養(yǎng)胃顆粒是否具有鎮(zhèn)痛、抗炎作用。方法:1.采用幽門結(jié)扎法模擬大鼠胃潰瘍模型,分組及給藥:將60只大鼠按體重隨機分為空白組、模型組、陽性對照藥組(三九胃泰顆粒,3.6g· kg-1)和白金養(yǎng)胃顆粒高、中、低劑量組(2.20,1.10,0.55g· kg-1),每組10只。每天給藥1次,連續(xù)給藥5天。2.模型制備:采用大鼠幽門結(jié)扎胃潰瘍實驗法[4]。首先各組鼠需禁食18h,但不禁水,把鼠放到空籠子中,以控制大鼠餓極吃食自己的糞便、墊料,18h后用乙醚麻醉,麻醉后使把大鼠固定在鼠板上,用碘伏或酒精消毒老鼠皮膚,沿腹中線(自劍突下緣)切開老鼠腹壁(注意切開大鼠皮膚時要先輕后重),切口約為2.5cm,用止血鉗撐開腹膜,用手指輕輕推左側(cè)肋緣,向上使胃暴露于視線中,找出胃和十二指腸交界處,在胃和十二指腸連接處穿線(線需事先放在酒精里消毒),切記勿傷周圍大血管及毛細血管,然后將幽門結(jié)扎,結(jié)扎后縫合切口,縫合過程中用阿司匹林和酒精消毒,紗布包扎,之后將各組動物均經(jīng)直腸給藥1次,放回鼠籠中,禁食不禁水。15 h后乙醚麻醉大鼠,并于頸總動脈取血[52]。沿切口打開腹腔,沿胃大彎剪開胃部,摘取全胃放到盛有酒精的燒杯中,觀察并測量胃潰瘍情況,測量胃潰瘍點,胃黏膜的損傷長度,將每只大鼠所有的損傷長度加和作為該鼠胃潰瘍指數(shù),并給每個粘膜損傷的程度評分,計算每只鼠所有損傷評分的加和,用"分"表示胃潰瘍指數(shù)[]。觀察白金養(yǎng)胃顆粒對減少大鼠潰瘍作用、抑制潰瘍指數(shù)的作用;并監(jiān)測各組中大鼠的胃液量、胃液酸度,血清中的胃動素、胃泌素含量,以及前列腺素E2(PGE2)、白介素-1(IL-1)、白介素-7(IL-7)的水平,表皮生長因子(EGF)、一氧化氮(NO)的含量。并檢測胃組織中生長抑素(SS)水平、丙二醛(MDA)、H+-K+-ATP酶、超氧化物歧化酶(SOD)含量的變化程度。小鼠鎮(zhèn)痛實驗中(選用熱板法),實驗前于熱板儀上測定痛閡值(第1次添后足的時間作為指標),重復(fù)測取2次正常痛閾值作為給藥前痛閾值[52]。選擇水域的水溫在(55℃ ± 0.5℃)時的痛閾值在5~30s內(nèi)的雌性小鼠50只,分組給藥1 h后開始測小鼠痛閾值,按標號分組的小鼠計好痛閾值,之后4h每隔1h測各小鼠的痛閾值。記錄好小鼠給藥前后痛閾值的大小,保存好完整原始實驗數(shù)據(jù)。采用小鼠耳腫脹法進行小鼠抗炎實驗,60只小鼠,雌雄參半,按體重隨機分,每組10只小鼠,一共分為5組,分成陽性對照藥,也就是三九胃泰組(5.20 g · kg-1),空白組、白金養(yǎng)胃顆粒高、中、低劑量組(3.00,1.5,0.75 g · kg-1),每天給藥1次,給藥1天。分組給藥后于末次給藥1 h后,在每只小鼠右耳廓(耳朵兩面)涂抹0.02mL二甲苯,以致炎癥發(fā)生。并在4h后處死小鼠(采用脫頸法),并沿耳廓線剪下兩耳,用8mm的打孔器分別在同一部位打下兩耳片,并用分析天平稱其重量,計算每只小鼠右耳片的質(zhì)量減去左耳片的質(zhì)量,就是腫脹程度,并計算抑制率[54]。結(jié)論:藥效學(xué)研究顯示,在大鼠幽門結(jié)扎型胃潰瘍實驗中,與模型組比較,白金養(yǎng)胃顆粒三個劑量組均能夠顯著減小大鼠胃潰瘍面積和大鼠胃潰瘍指數(shù),可明顯減少大鼠胃液量,降低胃酸濃度(p0.01),并且增加血清中PGE2水平,胃泌素、胃動素的的含量(p0.05)。在小鼠鎮(zhèn)痛、抗炎實驗中,白金養(yǎng)胃顆粒高、中劑量組均能夠降低二甲苯所致的小鼠耳腫脹程度(p0.05),白金養(yǎng)胃顆粒各給藥組均可不同程度提高小鼠痛閾值(p0.01,p0.05)。白金養(yǎng)胃顆粒可以治療幽門結(jié)扎型胃潰瘍疾病,并且白金養(yǎng)胃顆粒還具有鎮(zhèn)痛與抗炎作用。
