丙泊酚對(duì)高脂血癥大鼠空間學(xué)習(xí)記憶功能的影響及機(jī)制

【摘要】:目的:研究丙泊酚對(duì)高脂血癥大鼠空間學(xué)習(xí)記憶功能和海馬神經(jīng)元Tau蛋白磷酸化表達(dá)水平及老年斑沉積的影響。方法:將成模大鼠隨機(jī)分成4組:P10、P30、P75組分別腹腔注射丙泊酚(10,30,75mg/kg·d-1),對(duì)照組(G組)給予生理鹽水75mg/kg·d-1,連續(xù)注射5d。Morris水迷宮測(cè)試大鼠空間學(xué)習(xí)記憶功能。取海馬組織行剛果紅和免疫組織化學(xué)染色,ipwin32軟件分析Tau蛋白磷酸化表達(dá)陽(yáng)性細(xì)胞平均光密度值。結(jié)果:定位航行實(shí)驗(yàn)及附加實(shí)驗(yàn):P30、P75組逃避潛伏期較P10、G組明顯延長(zhǎng)(P0.05);空間搜索實(shí)驗(yàn):P30、P75組穿越平臺(tái)次數(shù)較P10、G組明顯減少(P0.05);免疫組織化學(xué)染色:P30、P75組平均光密度值與P10、G組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。剛果紅染色:P30、P75組可見(jiàn)老年斑沉積物。結(jié)論:鎮(zhèn)靜或麻醉劑量丙泊酚可損害高脂血癥大鼠空間學(xué)習(xí)記憶能力。其機(jī)制可能與海馬老年斑沉積增多和Tau蛋白過(guò)度磷酸化相關(guān)。
[Abstract]:Aim: to study the effects of propofol on spatial learning and memory function, phosphorylation of Tau protein in hippocampal neurons and plaque deposition in hyperlipidemia rats. Methods: the model rats were randomly divided into four groups: group P10, group P30, group P75 was injected with propofol (10, 30, 75 mg / kg 路d-1), and the control group (group G) was treated with saline 75mg/ kg 路d-1. The spatial learning and memory function of rats was measured by continuous injection of 5d.Morris water maze. The hippocampal tissues were stained with Congo red and immunohistochemistry, and the average optical density of Tau protein phosphorylation positive cells was analyzed by ipwin32 software. Results: the escape latency of P75 group was significantly longer than that of P10, and the escape latency of P75 group was significantly longer than that of P10 group (P 0.05), and the number of platform crossing in P75 group was significantly lower than that of P10 group (P 0.05), and the escape latency of P75 group was significantly longer than that of P10 group (P 0.05). The average optical density of P30 and P75 groups was significantly different from that of P10 and G groups (P 0.05). Congo red staining: P30, P75 group can see the deposit of old age spot. Conclusion: sedation or anaesthesia dose propofol can damage the spatial learning and memory ability of hyperlipidemia rats. The mechanism may be related to the increase of hippocampal plaque deposition and hyperphosphorylation of Tau protein.
