眼部不同途徑局部注射地塞米松對1型糖尿病大鼠血糖的影響

【摘要】:目的: 評價(jià)地塞米松球結(jié)膜下注射和球旁注射二種給藥途徑對1型糖尿病SD大鼠血糖的影響 方法: 選擇正常健康SD大鼠,雄性,鼠齡:2個(gè)月,,體重220±20g,分別在其腹腔注射鏈脲佐菌素(臨用前溶解在pH=4.0的0.1mmol/L檸檬酸緩沖液內(nèi),55mg/kg,1次),或注射等劑量的檸檬酸緩沖液,建立1型糖尿病SD大鼠模型。 選取造模成功的1型糖尿病SD大鼠(n=40)和正常SD大鼠(n=40)分別分為Ⅰ組1型糖尿病SD大鼠實(shí)驗(yàn)組和Ⅱ組正常SD大鼠對照組,將Ⅰ組1型糖尿病SD大鼠實(shí)驗(yàn)組隨機(jī)分為四組,每小組10只,ⅠA組為球結(jié)膜下注射地塞米松組,Ⅰa組為球結(jié)膜注射生理鹽水組;ⅠB組為球旁注射地塞米松組,Ⅰb組為球旁注射生理鹽水組。將Ⅱ組正常SD大鼠對照組隨機(jī)分為四組,每小組10只,ⅡA組為球結(jié)膜下注射地塞米松組,Ⅱa組為球結(jié)膜注射生理鹽水組;ⅡB組為球旁注射地塞米松組,Ⅱb組為球旁注射生理鹽水組。 大鼠禁食,不禁水12小時(shí)后,給予2%戊巴比妥鈉無菌液0.2ml/100g腹腔注射進(jìn)行麻醉,根據(jù)分組分別行球結(jié)膜下或球旁局部注射一次地塞米松0.1ml或注射等劑量的生理鹽水,注射開始后大鼠禁食不禁水,采尾靜脈收集血樣,利用血糖監(jiān)測儀(試紙)法測定在眼局部注射時(shí)及注射后3h、6h、12h、18h、24h時(shí)間點(diǎn)的血糖值。 結(jié)果: 利用鏈脲佐菌素腹腔注射,建立1型糖尿病SD大鼠模型,7天后測大鼠空腹血糖,造模成功1型糖尿病SD大鼠血糖水平(9.31±1.79)mmol/L,注射等劑量檸檬酸緩沖液后正常SD大鼠血糖水平(5.72±0.80)mmol/L,造模成功率為78%。 Ⅰ組1型糖尿病SD大鼠實(shí)驗(yàn)組,眼部不同途徑(球結(jié)膜下和球旁)注射地塞米松組血糖水平高于注射生理鹽水組,從注射后6h起,注射地塞米松組和注射生理鹽水組的血糖差異有統(tǒng)計(jì)學(xué)意義(P<0.05),組內(nèi)對比可知,注射地塞米松組和注射生理鹽水組均為3h的血糖水平最高,注射地塞米松組3h、12h、18h、24h,注射生理鹽水組3h、6h、12h、18h、24h時(shí)間點(diǎn)血糖水平,均與其各自注射前比較差異均有統(tǒng)計(jì)學(xué)意義(P<0.05)。 Ⅱ組正常SD大鼠對照組,球結(jié)膜下注射地塞米松組血糖水平均高于注射生理鹽水組,從注射后3h起,注射地塞米松組和注射生理鹽水組的血糖差異有統(tǒng)計(jì)學(xué)意義(P<0.05),組內(nèi)對比可知,注射地塞米松組3h、18h、24h,注射生理鹽水組3h、6h、12h、18h、24h時(shí)間點(diǎn)血糖水平,均與其各自注射前比較差異均有統(tǒng)計(jì)學(xué)意義(P<0.05)。球旁注射地塞米松組血糖水平均高于注射生理鹽水組,從注射后3h起,注射地塞米松組和注射生理鹽水組的血糖差異有統(tǒng)計(jì)學(xué)意義(P<0.05),組內(nèi)對比可知,注射地塞米松組3h、6h、18h、24h,注射生理鹽水組6h、12h、18h、24h時(shí)間點(diǎn)血糖水平,均與其各自注射前比較差異均有統(tǒng)計(jì)學(xué)意義(P<0.05)。 1型糖尿病SD大鼠實(shí)驗(yàn)組,無論注射地塞米松組或注射生理鹽水組,球結(jié)膜注射和球旁注射兩種方法的大鼠血糖值水平差異,均無統(tǒng)計(jì)學(xué)意義(P0.05)。 正常SD大鼠對照組,無論注射地塞米松組或注射生理鹽水組,球結(jié)膜注射和球旁注射兩種方法的大鼠血糖值水平差異,均無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論: 1、在1型糖尿病SD大鼠和正常SD大鼠,眼部球結(jié)膜下和球旁注射地塞米松均會造成大鼠血糖升高,1型糖尿病SD大鼠血糖升高更為明顯。提示地塞米松眼部的注射均會造成大鼠血糖升高。 2、在1型糖尿病SD大鼠和正常SD大鼠,眼部球結(jié)膜下和球旁注射生理鹽水后,1型糖尿病SD大鼠血糖升高,正常SD大鼠血糖不升高,提示局部注射操作引起大鼠應(yīng)激反應(yīng),可使1型糖尿病SD大鼠血糖升高。 3、球結(jié)膜下注射和球旁注射地塞米松,雖然造成1型糖尿病SD大鼠血糖升高,但血糖升高值無差異,提示這二種方法對大鼠血糖產(chǎn)生的影響沒有差異性。
