枸杞多糖對(duì)己烯雌酚暴露成年倉(cāng)鼠睪丸Caspase-8、Caspase-9及p53凋亡蛋白的影響

【摘要】:66只成年雄性倉(cāng)鼠隨機(jī)分為空白對(duì)照組(A)、DES組(B)、1 mg/kg LBP組(C)、10 mg/kg LBP組(D)、50mg/kg LBP組(E)及VC+VE組(F)。除對(duì)照組外,其余5組均皮下注射1mg/kg己烯雌酚,對(duì)照組注射等劑量的橄欖油,連續(xù)用藥7d,同時(shí)C、D、E組分別灌服1、10、50mg/kg的LBP,F組灌服100mg/kg的VC及200IU/kg的VE,A、B組灌服等劑量的生理鹽水。于末次給藥24h后,乙醚麻醉致死倉(cāng)鼠,取睪丸進(jìn)行免疫組化檢測(cè),確定Caspase-8、Caspase-9和p53的表達(dá)。結(jié)果顯示,通過(guò)免疫組化法檢測(cè)證實(shí),DES引起成年倉(cāng)鼠睪丸內(nèi)生精細(xì)胞大量凋亡,顯著提高Caspase-9的表達(dá)(P0.01);給予不同劑量的LBP后,Caspase-9及p53的表達(dá)呈劑量依賴性降低,以LBP高劑量組(50mg/kg)表達(dá)最低(P0.01),同時(shí)LBP高劑量組的Caspase-8陽(yáng)性表達(dá)與對(duì)照組相比差異不顯著。結(jié)果表明,枸杞多糖可以降低己烯雌酚所致成年倉(cāng)鼠生精細(xì)胞中凋亡蛋白Caspase-8、Caspase-9和p53的表達(dá),且以50mg/kg劑量作用最為顯著。
[Abstract]:66 adult male hamsters were randomly divided into blank control group, (A), DES group, (B), 1 mg/kg LBP group, (C), 10 mg/kg LBP group, (D), 50mg/kg LBP group (E) and VC VE group (F). In addition to the control group, the other five groups were subcutaneously injected with 1mg/kg diethylstilbestrol. The control group was injected with olive oil of the same dose for 7 days. Meanwhile, the control group was given the VC of 100mg/kg and the VE, of 200IU/kg of 50 mg / kg LBP,F. Group A and B were given the same dose of normal saline. 24 hours after the last administration, the testis were anesthetized to kill hamsters. The expressions of Caspase-8,Caspase-9 and p53 were determined by immunohistochemistry. The results showed that DES induced a large number of apoptosis of spermatogenic cells in the testis of adult hamsters and significantly increased the expression of Caspase-9 (P0.01). After different doses of LBP, the expression of Caspase-9 and p53 decreased in a dose-dependent manner, especially in the high dose LBP group (50mg/kg) (P0.01), and the expression of Caspase-8 in the LBP high dose group was not significantly different from that in the control group. The results showed that Lycium barbarum polysaccharides could decrease the expression of apoptotic protein Caspase-8,Caspase-9 and p53 in adult hamster spermatogenic cells induced by diethylstilbestrol, and 50mg/kg was the most significant one.
