線粒體保護(hù)劑對(duì)異氟醚麻醉老年小鼠海馬線粒體功能和突觸可塑性的影響

【摘要】:目的觀察線粒體保護(hù)劑SS-31對(duì)異氟醚麻醉老年小鼠線粒體功能和突觸可塑性的影響。方法老年雄性C57BL/6小鼠48只隨機(jī)均分為四組(n=12):氧氣+生理鹽水組(CN組)、氧氣+SS-31組(CS組)、異氟醚+生理鹽水組(IN組)和異氟醚+SS-31組(IS組)。根據(jù)分組,于氧氣或異氟醚吸入前30min腹腔注射等容生理鹽水(CN、IN組)或SS-31(5mg/kg,CS、IS組),于氣體吸入2h后取小鼠海馬組織。提取線粒體檢測(cè)電子傳遞鏈復(fù)合物Ⅰ~Ⅳ酶活性、三磷酸腺苷(ATP)和線粒體膜電位(MMP)水平,組織勻漿檢測(cè)突觸可塑性相關(guān)蛋白突觸蛋白1(synapsin-1)、突觸后密度蛋白95(PSD-95)、NMDA受體2A(NMDAR2A)、NMDAR2B、鈣調(diào)蛋白依賴的蛋白激酶Ⅱα(CaMKⅡα)和CaMKⅡβ的含量。結(jié)果與IN組比較,IS組復(fù)合物Ⅰ酶活性、ATP和MMP水平升高,synapsin-1和PSD-95含量增加,NMDAR2B、CaMKⅡα和CaMKⅡβ含量降低(P0.05),而復(fù)合物Ⅱ~Ⅳ酶活性及NMDAR2A含量差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論線粒體保護(hù)劑SS-31可改善異氟醚麻醉所致老年小鼠海馬線粒體及神經(jīng)元突觸可塑性損傷。
