糖尿病大鼠腦額葉皮層血流量的經(jīng)時(shí)變化

【摘要】:目的探討糖尿病大鼠腦額葉皮層血流量的動(dòng)態(tài)變化,初步研究其與主要血管活性物質(zhì)變化的關(guān)系,為進(jìn)一步研究糖尿病認(rèn)知障礙以及中樞神經(jīng)系統(tǒng)功能變化提供前期基礎(chǔ)。方法 34只♂Wistar大鼠隨機(jī)分為空白對(duì)照組和糖尿病模型組。模型組腹腔注射鏈脲佐菌素(STZ)60 mg·kg-1復(fù)制糖尿病模型,葡萄糖氧化酶法測(cè)定72 h空腹血糖≥16.7 mmol·L~(-1)作為成模標(biāo)準(zhǔn),血糖儀連續(xù)動(dòng)態(tài)測(cè)定FBG,Morris水迷宮檢測(cè)糖尿病大鼠學(xué)習(xí)記憶能力,雙通道激光多普勒血流儀檢測(cè)大鼠90 d額葉皮層血流量。另取34只♂Wistar大鼠分組造模同前,連續(xù)動(dòng)態(tài)監(jiān)測(cè)造模d 0、7、14、21、28、35、42、56、75額葉皮層血流量;ELISA法檢測(cè)腦脊液中i NOS、c NOS和ET-1的濃度。結(jié)果 Morris水迷宮檢測(cè)顯示,糖尿病模型組大鼠從訓(xùn)練第6次逃避潛伏期明顯延長(zhǎng)(P0.05),額葉皮層血流量/100 g體重明顯降低(P0.05);額葉皮層血流量動(dòng)態(tài)監(jiān)測(cè)結(jié)果顯示,糖尿病大鼠造模d 21腦血流量/100 g體重下降(P0.05),并在相似水平持續(xù)到成模d 75(P0.01,P0.05);與對(duì)照組相比,模型組腦脊液中i NOS和c NOS濃度明顯升高(P0.05),ET-1濃度明顯下降(P0.01)。結(jié)論糖尿病認(rèn)知障礙大鼠額葉皮層血流量減少,提示腦血流量的減少可能是導(dǎo)致糖尿病發(fā)生認(rèn)知障礙的因素之一。糖尿病動(dòng)態(tài)腦血流量檢測(cè)顯示腦血流量減少發(fā)生在糖尿病早期,之后并無(wú)明顯繼續(xù)惡化。腦血流減少可能與NO、ET-1濃度變化無(wú)明顯關(guān)系,但腦血流的變化趨勢(shì)可能與二者變化有關(guān)。
