【摘要】：目的:探討腫節(jié)風(fēng)對(duì)60COγ射線照射后在小型豬放射性肺損傷中的防護(hù)作用和機(jī)制。方法:60頭雄性巴馬小型豬隨機(jī)分為正常組、照射組和腫節(jié)風(fēng)組,在麻醉?xiàng)l件下對(duì)動(dòng)物行右胸單次15 Gy的照射,正常組不予照射。腫節(jié)風(fēng)組于放療前1周開始經(jīng)口腔給予腫節(jié)風(fēng)配方顆粒溶液(30 mg·kg-1),正常組和單照組給予等量的生理鹽水,直至實(shí)驗(yàn)結(jié)束。在照射后4,8,12,24周,從3個(gè)組中分別隨機(jī)取5頭小型豬采集右肺組織,行羥脯氨酸測(cè)定檢測(cè)肺組織中膠原含量;免疫印跡檢測(cè)轉(zhuǎn)化生長(zhǎng)因子-β1(TGF-β1),基質(zhì)金屬蛋白酶-2(MMP-2)和MMP-9的蛋白表達(dá);明膠酶譜法測(cè)定MMP-2和MMP-9的酶活性。結(jié)果:在照射后不同時(shí)間點(diǎn),照射組中羥脯氨酸含量,TGF-β1,MMP-2和MMP-9的蛋白表達(dá),MMP-2,MMP-9的酶活性較正常組明顯升高(P0.05);與照射組比較,腫節(jié)風(fēng)組的羥脯氨酸含量,TGF-β1,MMP-2和MMP-9的蛋白表達(dá),MMP-2,MMP-9的酶活性明顯降低,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:腫節(jié)風(fēng)可通過降低TGF-β1,MMP-2和MMP-9的蛋白表達(dá)和后者的酶活性,減緩放射性肺纖維化的發(fā)展,從而發(fā)揮放射防護(hù)作用。
[Abstract]:Objective: to investigate the protective effect and mechanism of TJE on radiation induced lung injury in miniature pigs after 60CO 緯-irradiation. Methods: 60 male Bama miniature pigs were randomly divided into three groups: normal group, irradiation group and swollen Jiefeng group. The animals were exposed to the right chest for 15 Gy under anesthesia, but the normal group was not irradiated. One week before radiotherapy, the patients in the control group were given the same amount of normal saline solution in oral cavity (30 mg kg-1), and the control group and the single irradiation group were given the same amount of normal saline until the end of the experiment. At 24 weeks after irradiation, the right lung tissues were collected from 5 miniature pigs randomly, and the collagen content in lung tissue was measured by hydroxyproline assay, and the protein expression of transforming growth factor 尾 1 (TGF- 尾 1), matrix metalloproteinase-2 (MMP-2) and MMP-9 were detected by Western blotting. The enzyme activities of MMP-2 and MMP-9 were determined by gelatinase method. Results: at different time points after irradiation, the content of hydroxyproline, the protein expression of TGF- 尾 1, MMP-2 and MMP-9, and the activity of MMP-2,MMP-9 in the irradiated group were significantly higher than those in the normal group (P0.05). The content of hydroxyproline, the protein expression of TGF- 尾 1, MMP-2 and MMP-9, and the activity of MMP-2,MMP-9 were significantly decreased in the control group (P0.05). Conclusion: TJD can slow down the development of radiation-induced pulmonary fibrosis by decreasing the protein expression of TGF- 尾 _ 1-MMP-2 and MMP-9 and the enzyme activity of the latter, thus exerting the radiation-protective effect.
