缺氧缺血海馬組織中自噬相關(guān)蛋白Beclin-1和LC3的表達(dá)變化以及雷帕霉素對其表達(dá)的影響

【摘要】:目的觀察缺氧缺血性腦損傷(HIBD)新生大鼠在不同時間點(diǎn)海馬組織中自噬相關(guān)蛋白Beclin-1及LC3的表達(dá)變化,并探討雷帕霉素對上述蛋白表達(dá)的影響。方法將生后7 d的108只Sprague-Dawley大鼠隨機(jī)分為假手術(shù)組、HIBD組和雷帕霉素組,每組36只。采用改良Rice法建立HIBD模型;假手術(shù)組分離左側(cè)頸總動脈但不予結(jié)扎,不行低氧處理;雷帕霉素組在模型制作前1 h予腹腔注射雷帕霉素(0.5 mg/kg)。各組大鼠分別于模型制作結(jié)束后0、6、12、24、48、72 h時間點(diǎn)麻醉后斷頭取腦,采用Western blot法檢測大鼠海馬組織Beclin-1及LC3蛋白表達(dá)的變化。結(jié)果 HIBD組Beclin-1蛋白及雷帕霉素組Beclin-1、LC3-Ⅱ蛋白表達(dá)水平均從0 h起開始升高,24 h時達(dá)到峰值后回落;HIBD組LC3-Ⅱ蛋白表達(dá)水平從0 h起開始升高,12 h時達(dá)到峰值后回落。與假手術(shù)組相比,HIBD組及雷帕霉素組Beclin-1、LC3-Ⅱ蛋白表達(dá)水平在各時間點(diǎn)均顯著升高(P0.05);與HIBD組相比,除12 h時LC3-Ⅱ蛋白水平外,雷帕霉素組各時間點(diǎn)Beclin-1、LC3-Ⅱ蛋白表達(dá)水平均明顯升高(P0.05)。結(jié)論海馬組織細(xì)胞經(jīng)缺氧缺血后出現(xiàn)自噬激活,而雷帕霉素能增強(qiáng)自噬這一表達(dá)過程。
[Abstract]:Objective to observe the expression of autophagy associated protein (Beclin-1) and LC3 in hippocampal tissue of neonatal rats with hypoxic-ischemic brain injury (HIBD) at different time points and to explore the effect of rapamycin on the expression of these proteins. Methods 108 Sprague-Dawley rats were randomly divided into sham operation group (n = 36) and rapamycin group (n = 36). The HIBD model was established by modified Rice method, the left common carotid artery was separated without ligation in sham-operated group, and the rapamycin group was injected intraperitoneally with rapamycin (0.5 mg/kg) one hour before the model was made. The rats in each group were anesthetized after anesthesia at the end of the model, and the changes of Beclin-1 and LC3 protein expression in hippocampus were detected by Western blot method at the time point of 0 ~ (12) and 24 ~ (24) ~ 48 ~ 72 h after anaesthesia, and the changes of Beclin-1 and LC3 protein expression in hippocampus of rats were detected by Western blot method.
