NADPH氧化酶下調(diào)海馬內(nèi)微清蛋白表達在老年小鼠術(shù)后認知功能障礙中的作用

【摘要】:目的觀察海馬內(nèi)微清蛋白(PV)表達變化在老年小鼠術(shù)后認知功能障礙(POCD)中的作用,并探討磷酸酰胺腺嘌呤二核苷酸(NADPH)氧化酶是否參與其中。方法采用異氟醚麻醉+剖腹探查術(shù)建立POCD動物模型。雄性16月齡C57BL/6小鼠21只,隨機均分為三組(n=7):對照+溶劑組(C+V組)、異氟醚麻醉+剖腹探查+溶劑組(POCD+V組)、異氟醚麻醉+剖腹探查+夾竹桃麻素組(POCD+A組)。POCD+A組術(shù)后1h腹腔注射夾竹桃麻素5mg/kg,其后每天注射一次直至術(shù)后第7天;其余兩組注射等容量的溶劑。術(shù)后第6天行曠場實驗和條件性恐懼實驗訓(xùn)練,第7天在給予藥物或溶劑后2h行條件性恐懼測試。行為學(xué)測試后2h取小鼠海馬組織,采用Western blot檢測PV、gp91phox、p22phox及羥基壬烯酸(4-HNE)的表達變化。結(jié)果三組小鼠總探索路程和中央格停留時間差異無統(tǒng)計學(xué)意義。與C+V組比較,POCD+V組在場景性條件恐懼測試中僵直時間明顯縮短,海馬PV表達明顯下降,而gp91phox、p22phox和4-HNE表達明顯升高(P0.05);與POCD+V組比較,POCD+A組在場景性條件恐懼測試中僵直時間明顯延長,海馬PV表達明顯升高,gp91phox、p22phox和4-HNE表達明顯降低(P0.05)。結(jié)論海馬NADPH氧化酶活化介導(dǎo)PV表達降低可能參與老年小鼠POCD。
[Abstract]:Objective to investigate the role of microalbumin (PV) expression in postoperatively aged mice with cognitive dysfunction (POCD), and to explore whether (NADPH) oxidase of adenine dinucleotide is involved in it. Methods POCD animal model was established by isoflurane anesthesia. Twenty-one male 16-month-old C57BL/6 mice, They were randomly divided into three groups: control group (C V group), isoflurane anesthesia group (POCD V group), isoflurane anesthesia group (POCD A group). After 5 mg / kg, the rats were injected once a day until the 7th day after operation. The other two groups were injected with the same volume of solvent. The open field test and conditioned fear test were performed on the 6th day after operation, and the conditioned fear test was performed on the 7th day at 2 hours after the administration of the drug or solvent. Western blot was used to detect the expression of PVGp91phoxP22phox and hydroxylazenoic acid (4-HNE) in hippocampus of mice 2 h after behavioral test. Results there was no significant difference in total exploration distance and residence time between the three groups. Compared with C V group, POCD V group had significantly shorter stiffness time and lower hippocampal PV expression. Compared with the POCD V group, the expression of gp91phoxtrophox p22phox and 4-HNE was significantly increased (P0.05). Compared with the POCD V group, the rigidity time was significantly prolonged and the expression of p22phox and 4-HNE in hippocampus was significantly increased (P0.05). Conclusion the decrease of PV expression mediated by NADPH oxidase activation in hippocampus may be involved in POCD expression in aged mice.
