【摘要】：目的:探究益腎寧心中藥對環(huán)境污染物去氧乙烯基環(huán)乙烯(4-vinylcyclohexene diepoxide,VCD)所致卵巢功能不全模型大鼠的療效。方法:40只21日齡離乳大鼠,隨機(jī)分為佐劑對照組、模型組、坤泰膠囊低劑量組、坤泰膠囊中劑量組、坤泰膠囊高劑量組,每組8只。除佐劑對照組外的四組均給予15天腹腔注射濃度為80mg/kg VCD溶液(VCD溶于芝麻油中)2.5ml/kg · d,佐劑對照組腹腔注射15天同等劑量芝麻油2.5ml/kg·d。于第一次腹腔注射VCD時,坤泰膠囊低、中、高劑量組分別灌胃給藥45天。待VCD注射完后70±5天,根據(jù)陰道涂片情況,將持續(xù)處于動情間期或持續(xù)動情周期紊亂的大鼠麻醉取材,進(jìn)行檢測大鼠血清促卵泡生成素(Follicle-Stimulating Hormone,FSH)、促黃體生成素(leuteinizing hormone,.LH)、雌二醇(estradiol,E2)、抗苗勒管激素(anti-Mullerian hormone,AMH)等;取出卵巢、子宮進(jìn)行形態(tài)學(xué)觀察。結(jié)果:①與佐劑對照組相比,模型組大鼠血清FSH明顯增高且AMH、E2明顯降低(P均0.05),LH有所增高(P0.05);經(jīng)坤泰膠囊干預(yù)后,FSH水平顯著降低(P0.05),AMH水平明顯增加(P0.05);②模型組大鼠卵巢濕重較佐劑對照組減輕(P0.05),坤泰膠囊干預(yù)后,坤泰膠囊低劑量組卵巢濕重增加明顯(P0.05);③模型組大鼠儲備卵泡及成熟卵泡計數(shù)明顯減少(P均0.05),坤泰膠囊干預(yù)后,儲備卵泡及成熟卵泡計數(shù)明顯增加(P均0.05);④光鏡下,模型組子宮內(nèi)膜變薄,管壁子宮腺體數(shù)量明顯減少。經(jīng)坤泰膠囊干預(yù)后,內(nèi)膜有所增厚,腺體數(shù)量明顯增加。結(jié)論:1、成功構(gòu)建了由VCD所致大鼠卵巢功能不全的模型,具體表現(xiàn)為:血清性激素水平、卵巢濕重、卵泡數(shù)量、卵巢子宮形態(tài)均發(fā)生了改變;2、坤泰膠囊可對上述改變有明顯改善調(diào)節(jié)作用。
[Abstract]:Aim: to investigate the effect of Yishen Ningxin traditional Chinese medicine on ovarian dysfunction induced by environmental pollutant 4-vinylcyclohexene diepoxide (DEVCD) in rats. Methods Forty 21-day-old weaned rats were randomly divided into adjuvant control group, model group, low dose group, middle dose group and high dose group with 8 rats in each group. The four groups except the adjuvant control group were given intraperitoneal injection of 80mg/kg VCD solution (VCD soluble in sesame oil) 2.5ml/kg d for 15 days, while the adjuvant control group received the same dose of sesame oil 2.5ml/kg d for 15 days. At the first intraperitoneal injection of VCD, Kuntai capsule was given orally for 45 days in low, medium and high dose groups. After 70 鹵5 days of VCD injection, according to the vaginal smear, anesthetized rats with continuous estrous interphase or continuous estrous cycle disorder were anesthetized. The serum levels of Follicle-Stimulating, leuteinizing, estradiolol E 2 and anti-Mullerian hormone were detected, and the ovary was removed and the uterus was observed. Results compared with the adjuvant control group, In model group, serum FSH was significantly increased and AMH- E _ 2 was significantly decreased (P < 0.05), FSH level was significantly decreased (P0.05) after intervention of Kun Tai capsule (P0.05) and the wet weight of ovary was significantly decreased in model group (P0.05) than that in adjuvant control group (P0.05), and that in Kun Tai capsule was significantly lower than that in adjuvant control group (P0.05), and that in Kun Tai capsule group was significantly lower than that in adjuvant control group (P0.05). The wet weight of ovary increased significantly (P0.05) in the low dose group of Kun Tai capsule (P0.05). The number of reserve follicles and mature follicles in the model group decreased significantly (P 0.05). After the intervention of Kun Tai capsule, the number of reserve follicles and mature follicles increased significantly (all P 0.05) under light microscope. In the model group, the endometrium became thinner and the number of uterine glands on the wall decreased significantly. After the intervention of Kuntai capsule, the intima was thickened and the number of glands was obviously increased. Conclusion: 1, the rat model of ovarian dysfunction induced by VCD was successfully constructed. The specific manifestations were: serum sex hormone level, wet weight of ovary, number of follicles. The uterine morphology of the ovary has been changed and Kun Tai capsule can obviously improve and regulate the above changes.
