川芎嗪對(duì)脂多糖誘導(dǎo)肺損傷大鼠肺組織水通道蛋白1、5表達(dá)影響的研究

【摘要】:目的:初步探討川芎嗪(tetramethylpyrazine,TMP)對(duì)脂多糖(lipopolysaccharide,LPS)誘導(dǎo)肺損傷大鼠肺組織水通道蛋白(aquaporins,AQPs)1、5表達(dá)的調(diào)節(jié)作用。方法:健康雄性SPF級(jí)SD大鼠32只隨機(jī)分為正常對(duì)照組(C組)、TMP處理組(T組)、LPS刺激組(L組)、TMP治療組(LT組),每組8只。L組采用尾靜脈注射6 mg/kg LPS,T組給予TMP 100 mg/kg腹腔注射,LT組大鼠注射LPS 1 h后立即給予TMP 100 mg/kg腹腔注射。8 h后麻醉處死大鼠留取肺組織標(biāo)本。測定肺組織濕/干重比(wet/dry ratio,W/D),蘇木素-伊紅(HE)染色觀察肺組織病理形態(tài),實(shí)時(shí)熒光定量PCR(quantitative real-time PCR,q RT-PCR)檢測肺組織勻漿AQP1和AQP5的m RNA水平,免疫組化染色、Western blot檢測肺組織AQP1、AQP5蛋白表達(dá)。結(jié)果:肺組織切片HE染色光鏡下可見C組、T組肺組織結(jié)構(gòu)完整、清晰;L組肺泡結(jié)構(gòu)紊亂,彌漫性肺泡間隔增厚,肺泡腔內(nèi)有炎性細(xì)胞浸潤、出血和水腫,而LT組上述病理表現(xiàn)較L組減輕。L組平均肺組織濕/干重比值(5.577±0.243)明顯高于其余3組(P0.01);與C、T 2組(3.430±0.251、3.251±0.142)相比,LT組濕/干重比值(4.384±0.236)增加,差異存在統(tǒng)計(jì)學(xué)意義(P0.01)。q RT-PCR結(jié)果顯示T組AQP1、AQP5的RNA水平(1.462±0.080、1.267±0.063)高于C組(1.160±0.229、0.940±0.087),同時(shí),L組兩者轉(zhuǎn)錄水平下調(diào)(0.607±0.203、0.139±0.046),而LT組m RNA水平(0.848±0.071、0.314±0.134)高于L組(P0.05),免疫組化(C組:2.096±0.114、2.285±0.266;T組:3.253±0.149、3.790±0.308;L組:1.133±0.202、1.068±0.221;LT組:1.562±0.085、1.647±0.239)、Western blot結(jié)果(C組:1.443±0.103、1.278±0.061;T組:1.667±0.087、1.644±0.102;L組:0.974±0.090、0.529±0.100;LT組:1.179±0.051、0.805±0.082)與q RT-PCR一致(P0.05)。結(jié)論:川芎嗪可有效減輕脂多糖誘導(dǎo)急性呼吸窘迫綜合征的肺水腫,起到積極的保護(hù)作用,其機(jī)制可能與上調(diào)AQP1、AQP5的表達(dá)有關(guān)。
[Abstract]:Aim: to investigate the regulatory effect of tetramethylpyrazine (TMP) on the expression of aquaporins (AQPs) in lung tissue of rats with lung injury induced by lipopolysaccharide (LPS). Methods: Thirty-two healthy male Sprague-Dawley rats with SPF grade were randomly divided into two groups: normal control group (group C) treated with TMP (group T) and LPS stimulated group (group L) (LT group). Each group (n = 8) received intraperitoneal injection of TMP100 mg/kg with tail vein injection of 6 mg/kg LPST. The rats in LT group were given TMP100 mg/kg intraperitoneally immediately after injection of LPS-1.8h later, the rats were anesthetized to collect the lung tissue. Lung tissue wet / dry weight ratio (wet/dry / W / D), hematoxylin eosin (HE) staining were used to observe the histopathology of lung tissue, the mRNA levels of AQP1 and AQP5 in lung homogenate were detected by quantitative real-time PCR, and the expression of AQP1 AQP5 protein in lung tissue was detected by immunohistochemical staining. Results: under HE staining, the lung tissue structure of group C was intact, the alveolar structure of group L was disorder, diffuse alveolar septum thickened, inflammatory cell infiltration, hemorrhage and edema were found in alveolar cavity. The mean lung wet / dry weight ratio in LT group (5.577 鹵0.243) was significantly higher than that in the other three groups (P0.01), and the wet / dry weight ratio (4.384 鹵0.236) in LT group was significantly higher than that in Con T2 group (3.430 鹵0.251 鹵3.251 鹵0.142). 宸紓瀛樺湪緇熻瀛︽剰涔,
