藿樸夏苓湯干預(yù)濕熱環(huán)境下高脂血癥大鼠體脂代謝的實驗研究

【摘要】:目的: 建立并觀察在濕熱環(huán)境條件下,高脂飼料喂飼的方法建立的高脂血癥大鼠模型,探討在相同濕熱環(huán)境下高脂血癥大鼠總膽固醇(TC)、總甘油三酯(TG)、低密度脂蛋白膽固醇(LDL-C)、高密度脂蛋白膽固醇(HDL-C)及腎周和睪丸周圍脂肪墊的變化。以藿樸夏苓湯(清熱去濕代表方劑之一)對模型大鼠進(jìn)行干預(yù),探討該方對濕熱環(huán)境下高脂血癥大鼠的降脂化濁作用機(jī)理,并與活血祛瘀代表方中成藥血脂康(主要成分:紅曲,功能活血降脂)以及西藥降脂藥物立普妥(阿托伐他汀鈣)進(jìn)行比較,觀察各組實驗間血脂水平及體內(nèi)臟器周圍脂肪的差異。 方法: 本實驗采用SD雄性大鼠,隨機(jī)分為7組,正常大鼠模型組、單純濕熱環(huán)境組、單純高脂飲食組、濕熱環(huán)境+高脂飲食組(模型組)、藿樸夏苓湯組、血脂康組、立普妥組,每組8只。除正常組、單純高脂飲食組外,其余5組均模擬嶺南地區(qū)氣溫的環(huán)境,每日置于溫度(36±2)℃,相對濕度為70-90%的人工氣候箱內(nèi)6小時。除正常組與單純濕熱環(huán)境組持續(xù)以普通飼料喂養(yǎng),其余5組,均以(76.7%的基礎(chǔ)飼料+3%的膽固醇粉+10%的豬油+0.3%的豬膽鹽+5%蛋黃粉+5%奶粉)高脂飼料喂食,連續(xù)喂養(yǎng)6周,每日觀察大鼠活動情況、大便性狀及皮毛變化。藿樸夏苓湯組(0.418g/kg)與血脂康組(0.047g/kg),在喂養(yǎng)高脂飼料時即開始灌胃給藥干預(yù),均早晚2次給藥,連續(xù)給藥6周;立普妥組(1.8g/kg)在造模后第3周開始每日給藥1次,連續(xù)給藥3周。實驗結(jié)束前一天,禁食不禁水12小時,麻醉后解剖大鼠,心臟采血分別檢測血清膽固醇(TC)、總甘油三酯(TG)、低密度脂蛋白膽固醇(LDL-C)、高密度脂蛋白膽固醇(HDL-C),手術(shù)分離大鼠雙腎周及睪丸周圍脂肪墊稱重,統(tǒng)計實驗數(shù)據(jù)并分析。 結(jié)果: 1.濕熱環(huán)境+高脂飲食組(模型組)大鼠于造模約1周開始出現(xiàn)大便次數(shù)增多,糞質(zhì)稀爛,皮毛發(fā)黃無光澤,活動減少等癥狀,約3周時上述癥狀達(dá)到高峰。第6周實驗結(jié)束時檢測大鼠血清TC、TG、HDL-C、LDL-C水平較正常大鼠均明顯升高,統(tǒng)計數(shù)據(jù)差異明顯(P0.05);且模型鼠腎周及睪丸周圍脂肪墊增重較正常大鼠明顯(P0.05)。 2.單純濕熱環(huán)境下大鼠血清TC、TG、HDL-C、LDL-C水平較正常大鼠均有輕度升高,但統(tǒng)計數(shù)據(jù)差異不明顯(P0.05)。濕熱環(huán)境+高脂飲食組(模型組)大鼠血清TC、TG、HDL-C、LDL-C水平較單純高脂飲食大鼠有輕度升高,但統(tǒng)計數(shù)據(jù)差異不明顯(P0.05)。 3.與模型組比較,藿樸夏苓湯組大鼠血清TC、TG、HDL-C、LDL-C水平及體內(nèi)臟器(雙腎及睪丸)周圍脂肪墊質(zhì)量均明顯降低,且存在著明顯的差異性(P0.01),提示藿樸夏苓湯能夠在一定程度上干預(yù)濕熱環(huán)境下高脂血癥大鼠的血脂代謝,且以降低血清甘油三酯的水平最顯著。 4.藿樸夏苓湯組大鼠在血清TC、TG、HDL-C、LDL-C及體內(nèi)臟器(雙腎及睪丸)周圍脂肪墊質(zhì)量較血脂康組大鼠明顯降低,這表示藿樸夏苓湯在干預(yù)療效上優(yōu)于血脂康組。藿樸夏苓湯組與立普妥組大鼠,在血清TC、TG、HDL-C、LDL-C及脂肪墊質(zhì)量無明顯差異,提示藿樸夏苓湯與立普妥在該模型中的降脂效果大致相同。 結(jié)論: 1.本研究嘗試在濕熱環(huán)境下配合高脂飲食模擬嶺南地區(qū)高脂血癥的形成,對于構(gòu)建中醫(yī)病癥結(jié)合的高脂血癥動物造模具有一定參考價值。研究結(jié)果提示,在氣候濕熱的環(huán)境下,高脂飲食大鼠存在體內(nèi)脂肪代謝紊亂,且濕熱環(huán)境可能在一定程度上影響體脂代謝。 2.藿樸夏苓湯作為清熱化濕代表方之一,能夠明顯降低模型大鼠的血清TC、TG、 HDL-C、LDL-C的水平,較中成藥血脂康具有明顯優(yōu)勢。且其降低模型大鼠血清TC、TG、HDL-C、LDL-C作用與立普妥相近。
[Abstract]:Purpose :
A rat model of hyperlipidemia was established and observed under the condition of damp - heat environment and high - fat feed . The changes of total cholesterol ( TC ) , total triglyceride ( TG ) , low density lipoprotein cholesterol ( LDL - C ) , high density lipoprotein cholesterol ( HDL - C ) and perirenal and testicular fat pad were discussed .
Method :
In this experiment , SD male rats were randomly divided into 7 groups , normal rats model group , simple damp - heat environment group , simple high - fat diet group , damp - heat environment + high - fat diet group ( model group ) , epimedin decoction group , Xuezhikang group and Lipitor group .
Serum cholesterol ( TC ) , total triglyceride ( TG ) , low density lipoprotein cholesterol ( LDL - C ) , high density lipoprotein cholesterol ( HDL - C ) , operation - isolated rat double - kidney week and peritesticular fat pad were weighed and statistically analyzed .
Results :
1 . At the end of the 6th week experiment , the levels of TC , TG , HDL - C , LDL - C in the serum TC , TG , HDL - C and LDL - C of rats were significantly higher than those in normal rats ( P0.05 ) .
The weight of fat pad in the perirenal and testis of the model rats was higher than that of the normal rats ( P0.05 ) .
2 . The levels of TC , TG , HDL - C and LDL - C in serum of rats were slightly higher than those in normal rats ( P0.05 ) . The levels of TC , TG , HDL - C , LDL - C in serum TC , TG , HDL - C and LDL - C in rats with damp - heat environment and high - fat diet group ( model group ) were slightly higher than those in normal rats , but the statistical difference was not significant ( P0.05 ) .
3 . Compared with model group , the serum TC , TG , HDL - C , LDL - C level and fat pad quality of internal organs ( double kidney and testis ) decreased significantly , and there was a significant difference ( P0.01 ) .
4 . The quality of fat pad in serum TC , TG , HDL - C , LDL - C and internal organs ( double kidney and testis ) was significantly lower than those in Xuezhikang group .
Conclusion :
1 . The study tried to simulate the formation of hyperlipemia in Ling Nan region by combining high fat diet with high fat diet under the humid and hot environment . The results suggested that in the environment of humid and hot climate , the fat metabolism in vivo was disturbed by high fat diet rats , and the damp - heat environment could affect body fat metabolism to some extent .
2 . As one of the representatives of heat - clearing and dampness - conditioning , the serum TC , TG , HDL - C , LDL - C of the model rats can be obviously reduced , and the levels of TC , TG , HDL - C and LDL - C in the model rats can be obviously reduced , and the serum TC , TG , HDL - C and LDL - C of the model rats are reduced to be similar to that of Lipitor .
