腦室內注射orexin-A和orexin-B對下丘腦組胺釋放的影響

【摘要】:目的觀察腦室內微注射orexin-A和orexin-B對下丘腦組胺釋放的影響,為orexins促麻醉覺醒的機制提供新的解釋。方法成年雄性SD大鼠隨機分為三組:生理鹽水組、orexin-A組和orexin-B組。在立體定位儀下將微透析管置入下丘腦結節(jié)乳頭體核區(qū)。實驗分為兩部分:先收集腦室內分別注射orexin-A、orexin-B 1nmol和5nmol后1、2h和3h的微透析液(n=5),采用高效液相色譜法(HPLC)檢測微透析液中組胺含量的時程變化。1周后,各組大鼠分別注入不同劑量的orexin-A和orexin-B(10、15nmol和20nmol,n=5),收集微注射后1h的透析液,檢測組胺釋放的劑量效應。在立體定位儀下結節(jié)乳頭體核置入微注射管,1周后給予1.4%異氟醚(1 MAC)麻醉30min,各組大鼠分別微注射生理鹽水、orexin-A和orexin-B 0.3μl(n=6),記錄翻正反射恢復時間。結果與生理鹽水組比較,腦室內微注射orexin-A 1nmol 2h后下丘腦組胺含量明顯增加,但同樣劑量的orexin-B無明顯作用,注射orexin-A和orexin-B 5nmol后2、3h組胺含量均明顯增加(P0.01)。腦室內微注射10nmol、15nmol和20nmol的orexin-A和orexin-B后1h組胺含量明顯高于生理鹽水,且20nmol含量最高(P0.05)。結節(jié)乳頭體核微注射orexin-A后翻正反射恢復時間明顯短于生理鹽水(P0.01),微注射orexin-B后無明顯作用。結論腦室內微注射orexin-A和orexin-B均可促進下丘腦組胺遞質的釋放,但orexin-A的作用更強。結節(jié)乳頭體核微注射orexin-A促進異氟醚麻醉的覺醒。
[Abstract]:Objective to investigate the effects of intraventricular microinjection of orexin-A and orexin-B on histamine release from hypothalamus and to provide a new explanation for the mechanism of anesthesia arousal induced by orexins. Methods Adult male SD rats were randomly divided into three groups: normal saline group, orexin-A group and orexin-B group. The microdialysis tube was placed into the nucleus of the papillary body of the hypothalamic tubercle under the stereotactic locator. The experiment was divided into two parts: after intraventricular injection of orexin-Anoorexin-B 1nmol and 5nmol, the microdialysis solution was collected for 1h and 3h, respectively. The histamine content in microdialysis solution was measured by HPLC for 1. 1 weeks. Rats in each group were injected with different doses of orexin-A and orexin-BX 10nmol and 20nmol / nmol respectively. Dialysate was collected at 1 hour after microinjection to detect the dose effect of histamine release. Rats were anesthetized with 1.4% isoflurane 1 Macl for 30 min after microinjection of papillary body nucleus in tuberculous nucleus under stereotactic locator. Rats in each group were given physiological saline troxin-A and orexin-B 0.3 渭 l / L for 30 min, respectively. The recovery time of positive inversion reflex was recorded. Results compared with saline group, the content of histamine in hypothalamus increased significantly after intraventricular microinjection of orexin-A 1nmol for 2 h, but the same dose of orexin-B had no obvious effect. The content of histamine increased significantly at 2h after injection of orexin-A and orexin-B 5nmol. The content of histamine in orexin-A and orexin-B at 1 h after microinjection of 10 nmol and 15 nmol and 20nmol was significantly higher than that of normal saline, and the highest content of 20nmol was P0.05. The recovery time of positive inversion reflex after microinjection of orexin-A was significantly shorter than that of P0.01A, but it had no effect after microinjection of orexin-B. Conclusion Intraventricular microinjection of orexin-A and orexin-B can promote the release of histamine transmitters in hypothalamus, but orexin-A is more effective. Microinjection of orexin-A into nodular papillary nucleus promotes the arousal of isoflurane anesthesia.
