清肺解毒飲對肺炎鏈球菌肺炎、肺炎克雷伯菌肺炎兩種老齡肺炎大鼠肺組織TNF-α、IFN-γ、IL-6及IL-10含量變化的

【摘要】:目的:通過建立肺炎鏈球菌與肺炎克雷伯菌兩種老齡大鼠的肺炎模型,運(yùn)用清肺解毒飲治療兩種老齡肺炎大鼠后,觀察其肺組織內(nèi)炎癥因子TNF-α,IFN-γ,IL-6,IL-10含量的變化,比較給藥前后肺組織內(nèi)炎癥因子指標(biāo)的變化,給清肺解毒飲治療兩種細(xì)菌性肺炎提供實驗依據(jù)。 方法:選用SD老齡大鼠20-22月齡,體重400g±50g,將140只(SPF級)SD老齡雄性大鼠隨機(jī)分為7組,分別為正常組、肺炎鏈球菌老齡大鼠模型組、肺炎克雷伯菌老齡大鼠模型組及兩種肺炎模型治療的清肺解毒飲高、低劑量組(分別簡稱肺鏈高劑量組、肺鏈低劑量組和肺克高劑量組、肺克低劑量組)。每組均20只,適應(yīng)性喂養(yǎng)一周,參照Yershov等方法,分別將肺炎鏈球菌、肺炎克雷伯菌氣管內(nèi)注入。將腹腔注射10%水合氯醛0.35mL/100g使大鼠麻醉,并經(jīng)上中切牙點懸吊,用一接注射器的鈍圓針頭經(jīng)口插入氣管,分別滴入40μL約含有107CFU處于對數(shù)生長期的肺炎鏈球菌、肺炎克雷伯菌混懸液(在滴入時堵住鼻孔),使接種物入肺,急性肺炎很快誘導(dǎo)成功。 按上述方法造模后灌胃給藥,每組給藥劑量均根據(jù)大鼠體重按成人一日劑量折算[按公式大鼠等效劑量(mg/kg)=折算系數(shù)×成人劑量(mg/kg)]計算出大鼠等效劑量:中藥:8.19g/kg,中藥高、低劑量組分別給予中藥16.4、4.10g/(kg·d)灌胃,正常組和兩種老齡大鼠肺炎模型組分別灌以生理鹽水。連續(xù)給藥14天,然后將大鼠處死。以檢查大鼠胸片、實驗室檢測及免疫組化的改變。 運(yùn)用安徽中醫(yī)藥大學(xué)第一附屬醫(yī)院呼吸科臨床常用方劑清肺解毒飲治療后,觀察清肺解毒飲對兩種肺炎大鼠肺組織內(nèi)TNF-α,IFN-γ,,IL-6,IL-10含量的變化,比較給藥前后肺炎指標(biāo)的改變。 結(jié)果:ELISA分析結(jié)果顯示,肺炎鏈球菌肺炎大鼠經(jīng)治療14天后的高劑量組肺組織內(nèi)含有的TNF-α、IFN-γ、IL-6、IL-10明顯低于模型組,有統(tǒng)計學(xué)意義。肺炎克雷伯菌肺炎大鼠經(jīng)治療14天后的高劑量組肺組織內(nèi)含有的TNF-α、IFN-γ、IL-6、IL-10明顯低于模型組,有統(tǒng)計學(xué)意義。兩種肺炎大鼠的低劑量治療組與其相對應(yīng)的模型組之間無統(tǒng)計學(xué)意義。 結(jié)論:清肺解毒飲在有效劑量下對肺炎鏈球菌及肺炎克雷伯菌兩種肺炎大鼠有治療作用,對相關(guān)細(xì)胞因子有調(diào)節(jié)作用。
[Abstract]:Objective: to establish the pneumonia model of two kinds of aged rats of Streptococcus pneumoniae and Klebsiella pneumoniae, and to observe the changes of IL-10 content of TNF- 偽 IFN- 緯 IL-6 in lung tissue of two aged rats treated with Qingfei jiedu decoction. The changes of inflammatory factors in lung tissue before and after administration were compared to provide experimental evidence for Qingfei jiedu decoction in the treatment of two kinds of bacterial pneumonia. Methods: 140 aged SD rats aged 20 to 22 months and weighing 400 g 鹵50 g were randomly divided into 7 groups: normal group and streptococcus pneumoniae model group. The Qingfei jiedu decoction treated by Klebsiella pneumoniae in aged rat model group and two kinds of pneumonia models were high and low dose groups (respectively referred to as lung chain high dose group, lung chain low dose group and lung chain high dose group, lung Ke low dose group). Twenty rats in each group were fed adaptively for one week. According to Yershov method, streptococcus pneumoniae and Klebsiella pneumoniae were injected into trachea respectively. The rats were anesthetized by intraperitoneal injection of 10% chloral hydrate (0.35mL/100g), suspended from the upper incisor, and inserted into the trachea with a blunt round needle attached to a syringe. 40 渭 L of Streptococcus pneumoniae containing 107CFU in logarithmic growth phase was dripped into the trachea, respectively. Klebsiella pneumoniae suspension (clogging the nostril at the time of dripping, allowing the inoculum to enter the lung, the acute pneumonia is quickly induced. According to the above method, the dosage of each group was converted according to the adult daily dose according to the body weight of rats [according to the formula, the equivalent dose of rats was calculated by the formula: the equivalent dose of Chinese medicine was 8.19 g / kg 路kg ~ (-1), and the dose of Chinese herbal medicine was high, according to the formula, the equivalent dose was 8.19 g 路kg ~ (-1) 路kg ~ (-1), and the dose of each group was 8.19 g / kg 路kg ~ (-1). The low dose group was given 16.4 ~ 4.10 g / kg / d, and the normal group and two kinds of aged rat pneumonia model group were given normal saline respectively. Rats were killed after 14 days of continuous administration. The changes of chest radiographs, laboratory examination and immunohistochemistry in rats were examined. After treatment with Qingfei jiedu decoction, a commonly used prescription in Department of Respiratory, the first affiliated Hospital of Anhui University of traditional Chinese Medicine, the changes of TNF- 偽 IFN- 緯 -IFN- 緯 IL-6 IL-10 in lung tissue of two kinds of pneumonia rats were observed, and the changes of pneumonia indexes before and after administration were compared. Results the percentage of TNF- 偽 IFN- 緯 IL-6 IL-10 in lung tissue of the high dose group was significantly lower than that of the model group after 14 days of treatment, the results showed that the IL-10 level in the lung tissue of the rats treated with streptococcus pneumoniae was significantly lower than that in the model group. The TNF- 偽 IFN- 緯 -IFN- 緯 IL-6 IL-10 in lung tissue of high dose group after 14 days of treatment with Klebsiella pneumoniae was significantly lower than that of model group. There was no significant difference between the low-dose treatment group and the corresponding model group. Conclusion: Qingfei jiedu decoction has therapeutic effect on Streptococcus pneumoniae and Klebsiella pneumoniae pneumonia rats under the effective dosage, and it can regulate related cytokines.
