海藻糖對(duì)異氟烷誘導(dǎo)轉(zhuǎn)APP基因小鼠學(xué)習(xí)記憶功能的影響

【摘要】:目的探討吸入麻醉藥異氟烷對(duì)轉(zhuǎn)APP基因小鼠學(xué)習(xí)記憶功能的影響及海藻糖的干預(yù)保護(hù)作用。方法 60只12月齡轉(zhuǎn)APP小鼠隨機(jī)分為空白對(duì)照組(Control組)、異氟烷組(Iso組)、海藻糖組(Iso+Tre組),每組20只。Iso+Tre組和Iso組分別于麻醉前30 min腹腔注射海藻糖400μg/kg或等量生理鹽水,然后給予1.4%異氟烷吸入麻醉2 h,麻醉后24 h應(yīng)用Morris水迷宮測(cè)試小鼠學(xué)習(xí)記憶能力,麻醉后6、24 h殺鼠取腦,采用TUNEL染色觀察各組小鼠海馬區(qū)神經(jīng)元凋亡,應(yīng)用ELISA法檢測(cè)Aβ含量,Western印跡法檢測(cè)Bcl-2、Bax、Caspase-3的表達(dá)變化。結(jié)果與Control組相比,Iso組小鼠總體的平均逃避潛伏期明顯延長(zhǎng),空間探索時(shí)間明顯縮短(P0.01),海馬神經(jīng)元凋亡指數(shù)、Aβ含量、Bax/Bcl-2比值及Caspase-3表達(dá)均明顯增加(P0.01);Iso+Tre組與Iso組相比,逃避潛伏期縮短,空間探索時(shí)間延長(zhǎng)(P0.05),Aβ含量、Bax/Bcl-2比值及Caspase-3表達(dá)均明顯減少(P0.01)。結(jié)論海藻糖能夠改善異氟烷誘導(dǎo)轉(zhuǎn)APP基因小鼠學(xué)習(xí)記憶能力的損害,減少海馬神經(jīng)元的凋亡及Aβ的生成。
[Abstract]:Objective to investigate the effects of inhaled anesthetic isoflurane on learning and memory function of APP transgenic mice and the protective effect of trehalose. Methods Sixty 12-month-old APP mice were randomly divided into control group (n = 20), isoflurane group (n = 20) and Iso group (n = 20). 20 rats in each group were intraperitoneally injected with trehalose 400 渭 g/kg or the same amount of normal saline 30 min before anesthesia. Then the mice were given 1.4% isoflurane inhalation anesthesia for 2 hours, the learning and memory ability of mice was tested by Morris water maze 24 hours after anesthesia, and the rat brain was killed 6 hours after anesthesia. The apoptosis of hippocampal neurons in each group was observed by TUNEL staining. ELISA assay was used to detect the content of A 尾. Western blot was used to detect the expression of Bcl-2 and Baxon Caspase-3. Results compared with the Control group, the average escape latency was significantly prolonged, the spatial exploration time was significantly shortened, and the apoptotic index of hippocampal neurons, the ratio of Bax / Bcl-2 and the expression of Caspase-3 in the hippocampal neurons were significantly increased in the Iso Tre group compared with the Iso group. The escape latency was shortened and the spatial exploration time was prolonged. The ratio of Bax / Bcl-2 and the expression of Caspase-3 decreased significantly. Conclusion trehalose can improve the learning and memory ability of APP transgenic mice induced by isoflurane and decrease the apoptosis of hippocampal neurons and the production of A 尾.
