依托咪酯對(duì)大鼠空間學(xué)習(xí)記憶能力及海馬CA1區(qū)LTD的影響研究

【摘要】:目的:通過Morris水迷宮研究依托咪酯對(duì)于大鼠空間學(xué)習(xí)記憶能力的影響及采用全細(xì)胞膜片鉗并結(jié)合藥理學(xué)方法,以長(zhǎng)時(shí)程抑制(Long-term depression,LTD)為指標(biāo),研究依托咪酯對(duì)大鼠空間學(xué)習(xí)記憶能力及海馬CA1區(qū)長(zhǎng)時(shí)程抑制的影響。方法:給予大鼠依托咪酯(5mg/kg)腹腔注射作為實(shí)驗(yàn)組,給予脂肪乳劑腹腔注射作為溶劑對(duì)照組,給予生理鹽水腹腔注射作為空白對(duì)照組,然后應(yīng)用Morris水迷宮評(píng)價(jià)依托咪酯對(duì)大鼠空間學(xué)習(xí)記憶的影響。之后應(yīng)用全細(xì)胞電壓鉗技術(shù),觀察30μl依托咪酯(相當(dāng)于5μmol/L)脂肪乳劑對(duì)大鼠海馬CA1區(qū)神經(jīng)元LTD的影響以及依托咪酯影響LTD的可能機(jī)制。結(jié)果:給予大鼠依托咪酯腹腔注射可以影響大鼠的空間學(xué)習(xí)記憶能力。依托咪酯可誘導(dǎo)LTD成功,且LTD表達(dá)增強(qiáng);LTD的易化作用能被GABAA受體競(jìng)爭(zhēng)性拮抗劑荷包牡丹堿阻斷。結(jié)論:研究表明多次給大鼠腹腔注射依托咪酯重復(fù)實(shí)施麻醉或鎮(zhèn)靜可以影響其空間學(xué)習(xí)記憶能力,且依托咪酯麻醉可以增強(qiáng)LTD的表達(dá),其機(jī)制可能影響大鼠空間學(xué)習(xí)記憶能力。
[Abstract]:Objective: to study the effect of etomidate on spatial learning and memory ability of rats by Morris water maze and to use whole-cell patch clamp and pharmacological method to study the long term expression of etomidate. To study the effects of etomidate on spatial learning and memory ability and long term inhibition of hippocampal CA1 in rats. Methods: the rats were given 5 mg / kg etomidate intraperitoneally as experimental group, fat emulsion as solvent control group and saline as blank control group. Then Morris water maze was used to evaluate the effect of etomidate on spatial learning and memory in rats. Then the effects of 30 渭 l etomidate (equivalent to 5 渭 mol / L) fat emulsion on LTD in rat hippocampal CA1 neurons and the possible mechanism of etomidate affecting LTD were observed by whole-cell voltage clamp technique. Results: intraperitoneal injection of etomidate could affect the ability of spatial learning and memory in rats. Etomidate could induce the success of LTD, and the enhancement of LTD expression could be blocked by bicuculline, a competitive antagonist of GABAA receptor. Conclusion: repeated intraperitoneal injection of etomidate or sedation may affect the ability of spatial learning and memory in rats, and etomidate anesthesia can enhance the expression of LTD, and its mechanism may affect the ability of spatial learning and memory in rats.
