貝母辛對(duì)LPS致黏液高分泌小鼠MUC5AC和EGFR表達(dá)的影響

【摘要】:目的:探討脂多糖致黏液高分泌小鼠MUC5AC表達(dá)的影響,并初步探討其機(jī)制。方法:用乙醚麻醉小鼠,經(jīng)鼻腔滴注脂多糖(1 mg/5 ml磷酸緩沖鹽溶液,每只50μl),構(gòu)建小鼠黏液高分泌模型。將小鼠隨機(jī)分為對(duì)照組、脂多糖組和2mg/kg、4mg/kg、6mg/kg劑量貝母辛組。HE染色觀察各組小鼠肺組織病理學(xué)改變、實(shí)時(shí)定量PCR法檢測(cè)MUC5AC mRNA表達(dá)、免疫組織化學(xué)法檢測(cè)MUC5AC蛋白表達(dá)及表皮生長(zhǎng)因子受體蛋白表達(dá)。結(jié)果:脂多糖組肺組織病理學(xué)顯示:氣道上皮脫落,氣道內(nèi)有粘液分泌,肺泡結(jié)構(gòu)呈實(shí)變,有大量炎癥細(xì)胞滲出,貝母辛治療組上述變化明顯減輕,且具有劑量依賴性。貝母辛(2mg/kg、4mg/kg、6mg/kg)均可降低脂多糖致黏液高分泌小鼠MUC5AC mRNA表達(dá),與脂多糖組相比,均有顯著的統(tǒng)計(jì)學(xué)意義,具有劑量依賴性。免疫組織化學(xué)法檢測(cè)MUC5AC蛋白,正常小鼠氣道上皮僅有少量的MUC5AC蛋白表達(dá),模型組明顯高于空白組,貝母辛可下調(diào)脂多糖誘導(dǎo)的MUC5AC表達(dá),具有劑量依賴性。貝母辛可下調(diào)脂多糖誘導(dǎo)的表皮生長(zhǎng)因子受體蛋白表達(dá),具有劑量依賴性。結(jié)論:貝母辛可降低脂多糖致黏液高分泌小鼠MUC5AC表達(dá),其機(jī)制可能與表皮生長(zhǎng)因子受體表達(dá)下調(diào)有關(guān)。
[Abstract]:Aim: to investigate the effect of lipopolysaccharide (LPS)-induced mucus hypersecretion on MUC5AC expression in mice and its mechanism. Methods: the mice were anesthetized with ether and injected with 1 mg/5 / ml phosphate buffer solution through nasal cavity to establish a mouse model of hypersecretion of mucus. The mice were randomly divided into control group, lipopolysaccharide group and 2 mg / kg / kg Beimuxin group. The pathological changes of lung tissue were observed by HE staining, and the expression of MUC5AC mRNA was detected by real-time quantitative PCR. The expression of MUC5AC protein and epidermal growth factor receptor protein were detected by immunohistochemistry. Results: in the lipopolysaccharide group, the lung histopathology showed that the airway epithelium was shedding, mucus secreted in the airway, the alveolar structure was solid, and a large number of inflammatory cells exudated. The above changes in the treatment group were obviously alleviated in a dose-dependent manner. The MUC5AC mRNA expression in mice induced by lipopolysaccharide was significantly lower than that in lipopolysaccharide group (P < 0.05). The expression of MUC5AC protein was detected by immunohistochemistry in normal mouse airway epithelium. The expression of MUC5AC protein in model group was significantly higher than that in blank group. Beimuxin could down-regulate the expression of MUC5AC induced by lipopolysaccharide in a dose-dependent manner. Beimuxin can down-regulate the expression of epidermal growth factor receptor protein induced by lipopolysaccharide in a dose-dependent manner. Conclusion: Beimuxin can reduce the expression of MUC5AC in mice induced by lipopolysaccharide and its mechanism may be related to the down-regulation of epidermal growth factor receptor expression.
