丙泊酚靜脈麻醉對(duì)大鼠水楊酸鈉排泄的影響

【摘要】:目的研究丙泊酚全身麻醉過程對(duì)水楊酸鈉排泄的影響及其機(jī)制。方法麻醉前,取雄性SD大鼠16只,腹腔注射1%水楊酸鈉80 mg·kg~(-1)后,收集尿液,記錄實(shí)際排尿時(shí)間和尿量。用光譜法測(cè)定尿液中水楊酸濃度,繪制尿藥濃度-時(shí)間曲線。經(jīng)3 d清洗期,上述大鼠尾靜脈注射丙泊酚150 mg·kg~(-1)后,立即腹腔注射水楊酸鈉,方法同前。繪制麻醉前與麻醉后尿藥濃度-時(shí)間曲線,代入藥代動(dòng)力學(xué)分析軟件3P97,計(jì)算尿藥代動(dòng)力學(xué)參數(shù),比較麻醉前與麻醉后水楊酸鈉排泄能力的變化。眶靜脈叢取血檢測(cè)血?dú)庵笜?biāo),并進(jìn)行血?dú)庵笜?biāo)與水楊酸累積排泄量相關(guān)分析。結(jié)果丙泊酚麻醉前與麻醉后,p H分別為7.44±0.04,7.37±0.06;二氧化碳分壓(PCO2)分別為(39.58±6.42),(47.71±6.80)mm Hg;氧分壓(PO2)分別為(88.25±6.80),(67.75±3.73)mm Hg;剩余堿(BE)分別為(1.20±1.05),(-5.08±1.88)mmol·L~(-1);最大排泄率(Rmax)分別為(0.62±0.15),(0.39±0.04)mg·h~(-1);Cmax分別為(1.43±0.23),(0.85±0.15)mg·kg~(-1);0~28 h排泄-時(shí)間曲線下面積(ERUC0-28)分別為(25.83±3.8),(17.35±2.01)mg·h·L~(-1),差異有統(tǒng)計(jì)學(xué)意義(均P0.05)。p H、PO2、BE與水楊酸累積排泄量呈明顯正相關(guān),相關(guān)系數(shù)(R2)分別為0.9758,0.9760,0.9398;而PCO2與水楊酸累積排泄量呈明顯反正相關(guān),R2為0.9023,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論丙泊酚靜脈麻醉能減慢大鼠水楊酸鈉排泄速度,與血?dú)庵笜?biāo)顯著相關(guān)。
[Abstract]:Objective to study the effect and mechanism of propofol general anesthesia on the excretion of sodium salicylate. Methods before anesthesia, 16 male Sprague-Dawley rats were injected intraperitoneally with 1% sodium salicylate (80 mg 路kg ~ (-1)). Urine was collected and the actual urination time and urine volume were recorded. The concentration of salicylic acid in urine was determined by spectral method. After 3 days of cleaning, sodium salicylate was injected intraperitoneally immediately after intravenous injection of propofol (150 mg 路kg ~ (-1)). The concentration-time curves of urinary drugs before and after anesthesia were drawn and the pharmacokinetic analysis software 3P97 was used to calculate the parameters of urinary pharmacokinetics and to compare the changes of excretion capacity of sodium salicylate before and after anesthesia.
