加味益氣活血湯對急性心肌缺血及高脂血癥模型小鼠的保護作用

【摘要】:目的:探討加味益氣活血湯(JIAWEIYIQIHUOXUE, JWYQ)對急性心肌缺血及高脂血癥模型小鼠的保護作用。 方法:①觀察氣管夾閉心肌缺血模型小鼠使用加味益氣活血方時心電消失時間的變化與影響:40只昆明雄性小鼠以每組10只,按體重隨機分為4組,分別為JWYQ大劑量組,JWYQ小劑量組,正常組,地奧組。正常組平時喂食普通小鼠飼料,其他組小鼠按灌胃量0.2ml/10g,每天灌胃JWYQ(39.OOg生藥/kg,19.50g生藥/kg),地奧心血康(0.195g/kg),連續(xù)20天后,在末次給藥45Min后對小鼠進行腹腔注射麻醉(20%烏拉坦aq,1.2g/kg)。麻醉完成后用小動脈夾夾閉小鼠氣管,觀察記錄心電消失時間。②50只昆明雄性小鼠以每組10只,按體重隨機分為5組,分別為JWYQ大劑量組,JWYQ、劑量組,正常組,對照組,地奧組。正常組平時喂食普通小鼠飼料,其他組小鼠按灌胃量.0.2ml/10g,每天灌胃JWYQ(39.OOg生藥/kg,19.50g生藥/kg),地奧心血康(0.195g/kg),對照組灌胃等量蒸餾水,連續(xù)20天后,在末次給藥45Min后除正常對照小組外,其余各小組均皮下注射Iso (0.015g/kg),建立急性心肌缺血模型,注射Iso注射液15min后,對5個小組小鼠置于密閉容器(250mi)中,分別記錄其存活時間。③50只昆明雄性小鼠以每組10只,按體重隨機分為5組,分別為JWYQ大劑量組,JWYQ小劑量組,正常組,對照組,辛伐他汀組。正常組平時喂食普通小鼠飼料,其它組均喂食高脂飼料。小鼠按灌胃量0.2ml/10g,每天灌胃JWYQ(39.OOg生藥/kg,19.50g生藥/kg),辛伐他汀(3mg/kg),連續(xù)20天后,在末次喂藥24h之后,各小組老鼠進行摘眼球取血,分離出血清,進行高脂血癥小鼠甘油三酯的對比測定。 結(jié)果:①中藥加味益氣活血方可以有效延長氣管夾閉實驗小鼠的心電消失時間。②中藥加味益氣活血方可以有效延長實驗小鼠注射Iso注射液誘發(fā)急性心肌缺血模型后在常壓密閉容器內(nèi)耐缺氧存活時間。③中藥加味益氣活血方可以有效降低高脂血癥實驗小鼠體內(nèi)甘油三酯。 使用大劑量的中藥加味益氣活血方延長氣管夾閉后心電消失時間的能力與注射Iso急性心肌缺血模型后耐缺氧存活時間的能力強于地奧心血康膠囊;且使用大劑量的中藥加味益氣活血方降低高脂血癥實驗小鼠體內(nèi)甘油三酯的能力也強于辛伐他汀膠囊。 結(jié)論:本次實驗研究表明加味中藥益氣活血方可以有效保護模型小鼠的急性心肌缺血,可以有效延長注射Iso誘發(fā)小鼠急性心肌缺血時密閉空間內(nèi)耐缺氧的存活時間,有效延長小鼠常壓密閉空間內(nèi)耐缺氧存活能力,能有效降低高脂血癥小鼠體內(nèi)甘油三酯(TG)。
[Abstract]:Objective: To investigate the protective effects of JIAWEIYIQIHUOXUE (JWYQ) on acute myocardial ischemia and hyperlipidemia mice.
Methods: (1) observe the change and influence of the time of electrocardio disappearance of the rat model of myocardial ischemia model with Jiawei Yiqi Huoxue Recipe: 40 male mice in Kunming were divided into 4 groups according to the weight of 10 rats. They were the large dose group, the JWYQ small dose group, the normal group and the Austrian group. The normal group was fed with ordinary mice feed and the other groups. Mice were treated with gastric perfusion 0.2ml/10g, JWYQ (39.OOg /kg, 19.50g raw drug /kg) and Diao Xin Xue Kang (0.195g/kg) every day. After 20 days of continuous delivery, the mice were anesthetized by intraperitoneal injection (20% uran AQ, 1.2g/kg) after the last administration of 45Min (20% uran AQ, 1.2g/kg). After the anesthesia was completed, the mouse trachea was clamped with a small artery clamp, and the time of disappearance of electrocardiogram was observed. 50 Kunming males were recorded. 10 rats in each group were divided into 5 groups according to their weight. They were JWYQ large dose group, JWYQ, dose group, normal group, control group, and Diao group. The normal group was fed with ordinary mice feed, the other groups were given.0.2ml/10g of gastric perfusion, JWYQ (/kg, 19.50g /kg), Diao Xin Xue Kang (0.195g/kg), and the control group gavage. After 20 days of distilled water, after the last dose of 45Min, the other groups were subcutaneously injected with Iso (0.015g/kg) to establish an acute myocardial ischemia model. After the injection of Iso injection 15min, 5 groups of mice were placed in the closed container (250mi), and the survival time was recorded respectively. (3) 50 Kunming male mice were treated with 10 mice in each group. The body weight was randomly divided into 5 groups: the JWYQ large dose group, the JWYQ small dose group, the normal group, the control group, the simvastatin group. The normal mice were fed with ordinary mice feed and the other groups were fed with high fat feed. The mice were fed with 0.2ml/10g, and were fed with JWYQ (/kg, 19.50g /kg) and simvastatin (3mg/kg) every day for the last 20 days, at the last feeding. After 24h, each group of mice took blood from the eyeballs and separated the bleeding to determine the triglyceride level in the hyperlipidemic mice.
Results: (1) the traditional Chinese medicine supplemented Yiqi Huoxue recipe can effectively prolong the time of electrocardiogram disappearance in the experimental mice. (2) the prescription of Supplementing Qi Yiqi and activating blood can effectively prolong the survival time of the experimental mice induced by the injection of Iso injection to the acute myocardial ischemia model in the normal pressure closed vessel. It can effectively reduce triglyceride in hyperlipidemic mice.
The ability of using a large dose of Chinese medicine plus Yiqi Huoxue recipe to prolong the time of disappearance of electrocardiogram after tracheal clamping and the ability to endure hypoxia survival time after injection of Iso acute myocardial ischemia model is stronger than that of dioxin Kang capsule, and the ability to reduce the ability of triglycerides in the experimental mice of hyperlipidemia with a large dose of traditional Chinese medicine plus Yiqi Huoxue recipe. It's better than Simvastatin Capsules.
Conclusion: the experimental study shows that the supplemented Chinese medicine Yiqi Huoxue recipe can effectively protect the acute myocardial ischemia of the model mice, and can effectively prolong the survival time of hypoxia tolerance in the closed space of the mice induced by Iso, effectively prolong the viability of the hypoxia tolerance in the confined space of the mice, and effectively reduce the high fat blood. Triglyceride (TG) in the body of the mice.
