替米沙坦對(duì)長期高糖高脂飲食誘導(dǎo)大鼠胰島PTP-1B表達(dá)的影響

【摘要】:目的:1.觀察替米沙坦對(duì)長期高糖高脂飲食誘導(dǎo)大鼠空腹血糖(FPG)、空腹胰島素(FINS)、甘油三酯(TG)、穩(wěn)態(tài)模型胰島素抵抗指數(shù)(HOMA-IR)的影響。2.觀察替米沙坦對(duì)長期高糖高脂飲食誘導(dǎo)大鼠胰島蛋白酪氨酸磷酸酶1B(PTP-1B)表達(dá)的影響,探討替米沙坦在改善胰島素抵抗方面可能的機(jī)制。方法:購買雄性SD大鼠,6-8周齡,30只,體重(180-200g),適應(yīng)性喂養(yǎng)1周,隨機(jī)分為3組:對(duì)照組(n=10)、高糖高脂組(n=10)、替米沙坦組(n=10)。對(duì)照組普通飼料喂養(yǎng),高糖高脂組和替米沙坦組高糖高脂喂養(yǎng),24周末,替米沙坦組給予灌胃替米沙坦10mg/kg/d劑量(生理鹽水溶解),干預(yù)12周。指標(biāo)檢測:替米沙坦干預(yù)12周后,大鼠禁食8—12小時(shí),稱重,腹腔注射烏拉坦麻醉,烏拉坦劑量4ml/kg,腹主動(dòng)脈采血1ml,血標(biāo)本經(jīng)低速離心機(jī)離心3000r/min,20min,離取血漿,立即送檢TG、FPG,葡萄糖氧化酶法測定FPG,全自動(dòng)生化分析儀測定TG。另血漿0.5ml于-70℃冰箱保存,同期測定FINS,胰島素ELISA試劑盒測定FINS。計(jì)算HOMA-IR=FPG×FINS/22.5。免疫組化分析:處死大鼠取血后放空血的方法,立即留取胰腺,把其置于10%中性福爾馬林濃度溶液固定24—48小時(shí),石蠟包埋,2um的厚度切片,行免疫組織化學(xué)染色,光學(xué)顯微鏡聯(lián)合美國aperio公司scanscope數(shù)字病理掃描系統(tǒng)測定胰島PTP-1B的含量。結(jié)果:1.替米沙坦干預(yù)12周后,與對(duì)照組相比,高糖高脂組FPG水平顯著增高(P0.05)。與高糖高脂組相比,替米沙坦組FPG水平顯著降低(P0.05)。2.替米沙坦干預(yù)12周后,與對(duì)照組相比,高糖高脂組血漿FINS水平顯著增高(P0.05)。與高糖高脂組相比,替米沙坦組血漿FINS水平顯著降低(P0.05)。3.替米沙坦干預(yù)12周后,與對(duì)照組相比,高糖高脂組血漿TG水平顯著增高(P0.05)。與高糖高脂組相比,替米沙坦組血漿TG水平顯著降低(P0.05)。4.替米沙坦干預(yù)12周后,與對(duì)照組相比,高糖高脂組HOMA-IR水平顯著增高(P0.05)。與高糖高脂組相比,替米沙坦組HOMA-IR水平顯著降低(P0.05)。5.替米沙坦干預(yù)12周后,與對(duì)照組相比,高糖高脂組PTP-1B含量顯著升高(P0.05)。與高糖高脂組相比,替米沙坦組PTP-1B含量顯著降低(P0.05)。結(jié)論:1.長期高糖高脂飲食誘導(dǎo)大鼠FPG、FINS、TG、HOMA-IR水平及胰島PTP-1B表達(dá)水平增高。2.替米沙坦干預(yù)12周后FPG、FINS、TG、HOMA-IR水平及胰島PTP-1B表達(dá)水平下降,推測替米沙坦改善胰島素抵抗與降低胰島PTP-1B蛋白表達(dá)有關(guān)。
[Abstract]:Purpose 1.To observe the effects of telmisartan on fasting blood glucose (FPG), fasting insulin (FINSN), triglyceride (TGN) and homeostasis index (HOMA-IRI) in rats induced by long-term high glucose and high fat diet.To observe the effect of telmisartan on the expression of islet protein tyrosine phosphatase (PTP-1B) in rats induced by high glucose and high fat diet for a long time, and to explore the possible mechanism of telmisartan in improving insulin resistance.Methods: 30 male Sprague-Dawley rats (6-8 weeks old) were randomly divided into three groups: control group (n = 10), high glucose and fat group (n = 10), telmisartan group (n = 10) and telmisartan group (n = 10).The control group was fed with common diet, the high-sugar and high-fat group and the telmisartan group were fed with high-sugar and high-fat diet for 24 weeks.
