【摘要】:目的:觀察參芪泄?jié)犸媽β阅I衰大鼠腎組織基質(zhì)金屬蛋白酶9(MMP-9)、基質(zhì)金屬蛋白酶抑制劑1(TIMP-1)基因表達(dá)的影響。方法:采用Yokozawa法復(fù)制慢性腎衰大鼠模型,隨機(jī)分為參芪泄?jié)犸嫺邉┝拷M、常規(guī)劑量組、氯沙坦組、模型組及空白組,高劑量組、常規(guī)劑量組、氯沙坦組分別予參芪泄?jié)犸嬎逡?0 m L/(kg·d)、參芪泄?jié)犸?0 m L/(kg·d)、氯沙坦9 mg/(kg·d),模型組與空白組大鼠均予純凈水10 m L/(kg·d),連續(xù)給藥8周,全部大鼠麻醉后腹腔采血,取左腎,ELLISA法測量血肌酐(Scr)、尿素氮(BUN)、丙氨酸轉(zhuǎn)氨酶(ALT)并采用熒光定量PCR檢測腎臟MMP-9、TIMP-1 mRNA的表達(dá)。結(jié)果:1高劑量組Scr水平明顯低于常規(guī)劑量組、氯沙坦組及模型組;常規(guī)劑量組與氯沙坦組無明顯差別,且均低于模型組;2高劑量組MMP-9 mRNA表達(dá)水平明顯高于常規(guī)劑量組、氯沙坦組及模型組;常規(guī)劑量組與氯沙坦組無明顯差別,且均高于模型組;高劑量組TIMP-1 mRNA表達(dá)水平明顯低于常規(guī)劑量組、氯沙坦組及模型組;常規(guī)劑量組與氯沙坦組無明顯差別,且均低于模型組;3各組ALT水平較空白組比較均無顯著差別。結(jié)論:參芪泄?jié)犸嬁赏ㄟ^調(diào)節(jié)MMP-9及TIMP-1表達(dá)來緩解慢性腎衰腎臟功能的惡化。
[Abstract]:Aim: to observe the effect of Shenqi Xiezhuo decoction (Shenqi Xiezhuo decoction) on the expression of matrix metalloproteinase 9 (MMP-9) and matrix metalloproteinase inhibitor 1 (TIMP 1) in renal tissue of rats with chronic renal failure (CRF).Methods: the rat model of chronic renal failure was established by Yokozawa method.
