虎杖苷對(duì)非酒精性脂肪肝保護(hù)作用的實(shí)驗(yàn)研究

【摘要】:目的:探討虎杖苷治療非酒精性脂肪肝的療效。方法:(1)造模:健康雄性SD大鼠47只,隨機(jī)分為5組:正常組(10只)、模型組(10只)、虎杖苷大劑量組(9只)、虎杖苷小劑量組(9只)和雙環(huán)醇組(9只)。正常組予標(biāo)準(zhǔn)飼料,其它4組給予高脂飼料;5組共喂養(yǎng)10周后,從正常組與模型組中各隨機(jī)挑選1只,取肝臟做病理切片,正常組大鼠肝臟切片作對(duì)照,證實(shí)模型組大鼠非酒精性脂肪肝的形成。(2)治療:正常組與模型組予以生理鹽水灌胃,5 m L/次;虎杖苷大、小劑量組分別以200 mg·kg-1·d-1,100 mg·kg-1·d-1的虎杖苷混懸液灌胃,雙環(huán)醇組以100 mg·kg-1·d-1的雙環(huán)醇混懸液灌胃。以上5組皆觀察4周,期間各組大鼠統(tǒng)一用標(biāo)準(zhǔn)飼料飼養(yǎng)。次日大鼠禁食12 h后,腹腔麻醉,1抽取腹主動(dòng)脈血5 m L,分離血清,檢測(cè)大鼠血清中肝功(丙氨酸氨基轉(zhuǎn)移酶ALT、天門冬氨酸氨基轉(zhuǎn)移酶AST)、血脂(總膽固醇TC、甘油三酯TG);2取肝組織,在大鼠肝臟的同一部位取4塊黃豆大小肝組織:一塊投多聚甲醛浸泡,為HE染色觀察肝臟病理學(xué)改變準(zhǔn)備。另兩塊裝Ep管,速投液氮。一份觀察油紅O染色觀察肝細(xì)胞內(nèi)脂質(zhì)沉積狀況。結(jié)果:1大鼠體質(zhì)量變化:造模第10周末,模型組、虎杖苷兩個(gè)劑量組和雙環(huán)醇組的大鼠體質(zhì)量明顯高于正常組(P0.05,0.01);治療1周后,與模型組相比虎杖苷大劑量組和雙環(huán)醇組體質(zhì)量開始出現(xiàn)降低(P0.05,0.01),治療第4周末,正常組、虎杖苷兩個(gè)劑量組及雙環(huán)醇組大鼠體質(zhì)量低于與模型組,差異具有統(tǒng)計(jì)學(xué)意義(分別對(duì)應(yīng)為P0.05,0.01),且虎杖苷大劑量對(duì)大鼠體質(zhì)量減輕效果與雙環(huán)醇組相當(dāng)(P0.05)。2肝功血脂的變化:治療4周后,與模型組相比,TC、TG在虎杖苷兩個(gè)劑量組、雙環(huán)醇組中均得到明顯改善(P0.05,0.01);⒄溶沾髣┝拷M對(duì)TG的降低作用優(yōu)于雙環(huán)醇組(P0.05)。3肝臟病理的變化:治療4周后,與模型組相比,肝細(xì)胞脂肪變、氣球樣變、小葉炎癥和門脈炎癥虎杖苷兩個(gè)劑量組、雙環(huán)醇組中均得到明顯改善。4肝臟油紅染色變化:治療4周后,與模型組相比,可見肝細(xì)胞脂肪含量在虎杖苷兩個(gè)劑量組、雙環(huán)醇組中均得到明顯改善。結(jié)論:虎杖苷對(duì)非酒精性脂肪肝具有較好療效,它可以減輕高脂飲食導(dǎo)致的非酒精性脂肪肝大鼠模型的體質(zhì)量增加、改善血脂肝功的異常、改善脂肪肝病理學(xué)改變和肝細(xì)胞脂質(zhì)沉積。
[Abstract]:Objective: to investigate the efficacy of polydatin in the treatment of non-alcoholic fatty liver. They were randomly divided into 5 groups: normal group (n = 10), model group (n = 10), large dose group (n = 9), polydatin group (n = 9) and bicyclic alcohol group (n = 9). One rat was randomly selected from the normal group and the model group, the liver was taken for pathological section, and the normal group was used as the control. It was confirmed that the formation of non-alcoholic fatty liver in the model group: normal group and model group were given normal saline for 5 mL / time, and the large and low dose groups were perfused with 200 mg kg-1 d-1100 mg kg-1 d -1 suspension of polydatin, respectively. The rats in the bicyclic alcohol group were fed with 100mg kg-1 d-1 bicyclic alcohol suspension for 4 weeks, during which the rats in each group were fed with standard diet. After fasting for 12 hours, the abdominal aorta blood was extracted from the rats under intraperitoneal anaesthesia for 5 mL and the serum was isolated. Liver function (alanine aminotransferase alt, aspartate aminotransferase ASTT), total cholesterol (TC) and triglyceride (TGG) were measured in serum of rats. Four pieces of soybean liver tissue were taken from the same part of the rat liver: one was soaked in paraformaldehyde to prepare for HE staining to observe the pathological changes of the liver, the other two were filled with EP tubes. A fraction of oil red O staining was used to observe the lipid deposition in hepatocytes. Results the body mass of rats at 1: 1 was changed at the end of the 10th week of model making, the model group, The body mass of the rats in the two dose groups of polydatin and bicyclol groups was significantly higher than that in the normal group (P0.050.01), and after one week of treatment, the body mass of the high-dose group and bicyclic alcohol group began to decrease compared with the model group, and the body mass of the rats in the high-dose group and bicyclic alcohol group began to decrease, and at the 4th week of treatment, the body mass of the rats in the normal group began to decrease. The body mass of rats in two dose groups of polydatin and bicyclol groups was lower than that in the model group. The difference was statistically significant (corresponding to P0.05P0.01G, respectively, and the effect of high-dose polydatin on reducing body mass in rats was equivalent to that in bicyclol group P0.050.2. the changes of lipids in liver function: after 4 weeks treatment, compared with the model group, TCTG was in the two dose groups of polydatin. The decrease of TG in the high-dose group of polydatin was better than that in the group of dicyclic alcohol: after 4 weeks of treatment, compared with the model group, the fatty and balloon changes of hepatocytes were observed. The changes of liver oil red staining were significantly improved in the two dose groups of polydatin and polydatin in lobular inflammation and portal inflammation. After 4 weeks of treatment, compared with the model group, the fatty content of hepatocytes was found to be in the two dose groups of polydatin. Conclusion: polydatin has a better effect on non-alcoholic fatty liver, it can reduce the increase of body weight and improve the abnormal lipids liver function in rats induced by high-fat diet. Improve fatty liver pathological changes and liver cell lipid deposition.
