【摘要】：[目的]：通過前列靈膠囊對慢性前列腺炎大鼠治療作用的實(shí)驗(yàn)研究,評價其治療效果,探討前列靈膠囊對慢性前列腺炎炎癥調(diào)控網(wǎng)絡(luò)及氧化應(yīng)激水平的影響,為治療慢性前列腺炎中藥的開發(fā)應(yīng)用奠定實(shí)驗(yàn)和理論基礎(chǔ)。 [方法]：將60只大鼠馴化喂養(yǎng)一周后,隨機(jī)分為6組,即空白組、陽性對照組、模型組、前列靈膠囊高劑量組、中劑量組、低劑量組。每組10只,各組用25%烏拉坦1gmg量,經(jīng)腹腔注射麻醉,無菌條件下取下腹正中切口,打開腹腔,提出膀胱及兩側(cè)精囊,暴露前列腺背葉。其中空白組分別經(jīng)前列腺雙側(cè)背葉注入無菌注射用水O.1ml,其余各組分別注入25%消痔靈注射液0.1ml,縫合后恢復(fù)1周。 給藥方法：空白組和模型組每日灌胃一次,每次灌胃與實(shí)驗(yàn)組等劑量的生理鹽水；陽性對照組(前列通瘀膠囊)：每只大鼠經(jīng)灌胃給予前列通瘀膠囊混懸液劑量相當(dāng)于60kg成人用量20倍；前列靈高、中、低劑量組：每組大鼠給予前列靈膠囊混懸液灌胃,劑量分別相當(dāng)于60kg成人用量20倍、10倍、5倍；均每日灌胃一次；連續(xù)給藥30天,每天觀察并記錄大鼠活動、體重及精神狀態(tài)變化,于末次給藥后24小時,稱每只大鼠的體重,每只大鼠采股動脈血3-5m1,離心冷藏,斷頭處死,摘除前列腺組織,前列腺組織用4%甲醛固定液固定。采用ELISA法分析各組大鼠血清中IL-6、ACP和SOD、MDA的含量。用S-P法檢測前列腺組織中GSH、GST、GSH-px含量,用電鏡觀察前列腺組織中亡因子Bax.Bcl-2的變化。 [結(jié)果]：前列靈膠囊可降低前列腺炎大鼠血清中IL-6.ACP.MDA和升高SOD的含量,以及升高慢性前列腺炎大鼠前列腺組織中GSH. GST.GSH-px的含量,前列靈膠囊還具有上調(diào)Bax蛋白含量和降低Bcl-2蛋白含量的作用。 [結(jié)論]：前列靈膠囊對大鼠慢性前列腺炎有明顯的治療作用,其作用機(jī)理與降低前列腺炎大鼠血清中IL-6、ACP、MDA和升高SOD含量有關(guān),以及升高慢性前列腺炎大鼠前列腺組織中GSH.GST. GSH-px的含量有關(guān),前列靈膠囊還具有上調(diào)Bax蛋白含量和降低Bcl-2蛋白含量的作用。
[Abstract]:[objective]: to evaluate the therapeutic effect of Qianlieling capsule on chronic prostatitis rats, and to explore the effect of Qianlieling capsule on inflammatory regulatory network and oxidative stress level in chronic prostatitis rats. To lay an experimental and theoretical foundation for the development and application of traditional Chinese medicine for the treatment of chronic prostatitis. [methods] Sixty rats were randomly divided into 6 groups: blank group, positive control group, model group, high dose group, middle dose group, low dose group, 10 rats in each group, and each group was given 25% uratan 1 g mg, and 10 rats in each group were randomly divided into 6 groups, namely the blank group, the positive control group, the model group, the high dose group of Qianlieling capsule, the middle dose group and the low dose group. Anesthetized by intraperitoneal injection, the median incision of the lower abdomen was taken under aseptic conditions, the abdominal cavity was opened, and the bladder and seminal vesicles were put forward. In the blank group, 0.1 ml of water was injected into the bilateral dorsal lobe of the prostate, and 0.1 ml of 25% Xiaozhiling injection was injected into the rest groups, and then recovered for 1 week after suture. Methods: the blank group and the model group were given intragastric once a day, and each time they were given the same dose of normal saline as the experimental group. The positive control group (Qianlie Tongyu capsule: the dosage of Qianlie Tongyu capsule suspension was 20 times that of 60 kg adult rats, the dosage of Qianlie Ling high, middle dose and low dose group: each group rats were given Qianliesling capsule suspension, the dosage of Qianlie Tongyu capsule suspension was 20 times as much as 60 kg adults, the rats of each group were given Qianlieling capsule suspension, The doses were 20 times as much as 10 times as much as 5 times as much as 60 kg of adults. They were given orally once a day. For 30 consecutive days, the rats' activities, body weight and mental state were observed and recorded every day, and the weight of each rat was weighed 24 hours after the last administration. The femoral arterial blood was collected from each rat from 3 to 5 m1.The rats were cryopreserved centrifugally, killed by decapitation, and the prostate tissue was removed. The contents of IL-6ACP and SDD in serum of rats in each group were analyzed by ELISA method. The GSH-px content in prostate tissue was detected by S-P method. The changes of Bax.Bcl-2 in prostate tissue were observed by electron microscope. [results]: Qianlieling capsule could decrease the content of IL-6.ACP.MDA and increase the content of SOD in serum of rats with prostatitis, and increase the content of GSH. GST.GSH-px in prostate tissue of rats with chronic prostatitis. Qianlieling capsule can also up-regulate the content of Bax protein and decrease the content of Bcl-2 protein. [conclusion]: Qianlieling capsule has obvious therapeutic effect on chronic prostatitis in rats, and its mechanism is related to the decrease of IL-6 ACP MDA and the increase of SOD content in serum of rats with prostatitis. The content of GSH.GST. GSH-px in prostatic tissue of rats with chronic prostatitis was increased, and the content of Bax protein was up-regulated and the content of Bcl-2 protein was decreased by Qianlieling capsule.
