七氟醚麻醉對老年小鼠空間認知能力以及海馬組織pCreb和Egr1表達的影響

【摘要】:目的探討七氟醚麻醉對老年小鼠空間認知能力以及海馬組織中磷酸化環(huán)腺苷酸應答元件結(jié)合蛋白(pCreb)和早期生長反應因子1(Egr1)表達的影響。方法將12只18月齡老年小鼠隨機分為麻醉組和對照組,每組各6只。麻醉組小鼠吸入3%七氟醚4 h,而對照組小鼠吸入100%氧氣4 h。水迷宮空間學習記憶能力測量在麻醉后48 h進行,獲得訓練結(jié)束后24 h進行空間探索試驗。空間探索試驗結(jié)束后15 min分離海馬組織,通過Western blotting和實時定量PCR方法檢測海馬組織Egr1、Creb和pCreb的表達。結(jié)果麻醉組小鼠第4、5、6日逃逸潛伏期顯著長于對照組(P0.05);在空間探索試驗中,對照組平臺象限時間、路程百分比顯著高于麻醉組(P0.01)。與對照組比較,麻醉組小鼠海馬組織Egr1的表達量顯著下降(P0.05);而兩組Creb的表達量無顯著差異(P0.05),但麻醉組磷酸化水平下降(P0.05)。結(jié)論七氟醚麻醉下調(diào)海馬組織Egr1表達和Creb磷酸化水平,影響老年小鼠空間認知能力。
[Abstract]:Objective to investigate the effects of sevoflurane anesthesia on spatial cognitive ability and the expression of phosphorylated cyclic adenylate responsive element binding protein (pCreb) and early growth response factor 1 (Egr1) in the hippocampus of aged mice. Rats were randomly divided into anesthetic group and control group. There were 6 mice in each group. The mice in the anesthesia group inhaled 3% sevoflurane for 4 hours, while the mice in the control group inhaled 100% oxygen for 4 hours. The spatial learning and memory ability of the water maze was measured 48 hours after anesthesia. The space exploration experiment was carried out 24 hours after the completion of the training. Hippocampal tissue was isolated from the hippocampus 15 min after the completion of the space exploration test. Western blotting and real-time quantitative PCR were used to detect the expression of Egr1C Creb and pCreb in hippocampal tissue. Results the escape latency of mice in the anesthetized group was significantly longer than that in the control group on 6th, and the time of platform quadrant in the control group was significantly longer than that in the control group. The percentage of distance was significantly higher than that of the anesthetic group (P 0.01). The expression of Egr1 in hippocampal tissue of anesthetized mice was significantly decreased (P 0.05), while the expression of Creb was not significantly different between the two groups, but the level of phosphorylation was decreased in anesthesia group. Conclusion sevoflurane anesthesia can down-regulate the expression of Egr1 and the level of Creb phosphorylation in hippocampus. The spatial cognitive ability of aged mice was affected.
