不同品系正常成年小鼠聽性腦干反應(yīng)特點(diǎn)

【摘要】:目的研究正常成年CBA、C57BL、昆明系和129系四種品系小鼠的聽性腦干反應(yīng)(auditory brainstem response,ABR),分析其ABR反應(yīng)閾和潛伏期,得到正常值范圍,標(biāo)準(zhǔn)化測(cè)試流程,為小鼠的聽覺相關(guān)研究提供參考。方法對(duì)6周齡正常CBA、C57BL、昆明系和129系四種品系小鼠各20只(20耳),完全麻醉后,進(jìn)行短聲(click)和短純音(tone burst,tb)ABR測(cè)試,觀察四種品系小鼠ABR各波的引出率,記錄引出率最高的波反應(yīng)閾值及各波潛伏期。結(jié)果 (1)在80dB SPL強(qiáng)度下,CBA、C57BL、昆明系和129系四種品系小鼠ABR各波引出率均為100%,CBA小鼠ABR波形分化最好;在30dB SPL強(qiáng)度下,除129小鼠外,其余3種小鼠波Ⅱ引出率最高,129小鼠在閾值附近50dB SPL強(qiáng)度下也是波Ⅱ引出率最高;129小鼠ABR反應(yīng)閾明顯高于其它三種小鼠,差異均有統(tǒng)計(jì)學(xué)意義(P均0.05);(2)四種品系小鼠的短純音聽覺敏感頻率為8、12、16kHz,其中對(duì)12kHz的短純音最敏感;(3)麻醉情況下,ABR各波潛伏期隨著測(cè)試時(shí)間延長(zhǎng)而延長(zhǎng),尤其波Ⅲ以后各波潛伏期延長(zhǎng)更明顯。80dB SPL強(qiáng)度下CBA、C57BL、昆明系、129系四種小鼠ABR波形在波Ⅰ前均可分化出SP(summating potential,SP)波,引出率分別為100%、90%、80%、50%,相同刺激條件下,SP波出現(xiàn)率高的小鼠其ABR反應(yīng)閾相對(duì)較低。結(jié)論麻醉狀態(tài)下,CBA、C57BL、昆明系和129系四種品系小鼠短聲及短純音ABR波Ⅱ引出率最高,故以波Ⅱ剛剛出現(xiàn)的刺激聲強(qiáng)度確定為反應(yīng)閾值;C57BL、昆明系和129系三種品系小鼠的ABR結(jié)果均沒有CBA小鼠穩(wěn)定,四種品系小鼠中CBA小鼠更適合用來(lái)建立聽覺功能相關(guān)研究的動(dòng)物模型;SP波可間接反映毛細(xì)胞功能。
[Abstract]:Objective to study the auditory brainstem response (ABR) of normal adult CBA C57BL, Kunming strain and 129-strain mice, and to analyze the threshold and latency of ABR response, and to obtain the normal range and standardized test procedure. Methods A total of 20 mice (20 ears from Kunming strain and 129 strain) were anesthetized to test the ABR. The extraction rate of ABR waves in four strains of mice was observed. Results at 80 dB SPL intensity, the highest threshold of wave response and the latency of each wave were recorded in Kunming strain and 129-strain mice, respectively. The extraction rates of ABR waves in Kunming and 129-strain mice were all the best, and at 30 dB SPL intensity, the ABR waveform differentiation was the best in Kunming strain and 129-strain mice, except for 129dB SPL intensity. Under the intensity of 50dB SPL near the threshold, the ABR reaction threshold of the other three kinds of mice was significantly higher than that of the other three kinds of mice, and the ABR reaction threshold of the other three mice was significantly higher than that of the other three kinds of mice. The difference was statistically significant (P < 0.05)) the frequency of short tone auditory sensitivity in four strains of mice was 812kHz and 16kHz, and the latency of ABR waves was prolonged with the prolongation of test time under anesthetized condition.
