【摘要】：目的研究槲皮素改善大鼠銅綠假單胞菌(Pesudomonas aeruginosa,PA)肺感染。方法 40只SPF級SD大鼠隨機(jī)分為5組,每組8只,分別為正常組、銅綠假單胞菌感染組、槲皮素組、左氧氟沙星組、左氧氟沙星聯(lián)用槲皮素組(聯(lián)用組),大鼠麻醉后支氣管注入銅綠假單胞菌。觀察大鼠肺組織病理變化,檢測肺組織IL-4、IFN-γ細(xì)胞因子含有量,轉(zhuǎn)錄因子T-bet、Gata-3的變化;用RT-PCR半定量法檢測IL-4、IFN-γ、T-bet、Gata-3 mRNA的表達(dá)。結(jié)果槲皮素組、左氧氟沙星組及聯(lián)用組肺組織IL-4含有量,IL-4、Gata-3 mRNA水平均較銅綠假單胞菌感染組降低,而IFN-γ、T-bet mRNA水平較銅綠假單胞菌感染組升高。結(jié)論槲皮素及左氧氟沙星能誘導(dǎo)大鼠機(jī)體由Th2型向Th1型分化,但兩者之間無協(xié)同作用。
[Abstract]:Objective to study the effects of quercetin on pulmonary infection of Pesudomonas aeruginosa in rats. Methods Forty SPF grade SD rats were randomly divided into 5 groups: normal group, Pseudomonas aeruginosa infection group, quercetin group and levofloxacin group. Pseudomonas aeruginosa was injected into the bronchus of rats after anesthesia with levofloxacin combined with quercetin. The pathological changes of lung tissue were observed, and the contents of IL-4 and IFN- 緯 cytokines and the changes of transcription factor T-beta Gata-3 in lung tissue were detected. RT-PCR semi-quantitative assay was used to detect the expression of IL-4 IFN- 緯 T-beta-Gata-3 mRNA. Results in quercetin group, levofloxacin group and combined group, the level of IL-4 Gata-3 mRNA in lung tissue was lower than that in Pseudomonas aeruginosa infection group. Conclusion quercetin and levofloxacin can induce the differentiation from Th2 type to Th1 type in rats, but there is no synergistic effect between them.
