七氟醚對(duì)新生大鼠下丘腦-垂體-腎上腺皮質(zhì)軸發(fā)育的影響

【摘要】:目的研究新生期七氟醚暴露對(duì)下丘腦-垂體-腎上腺皮質(zhì)(hypothalamic-pituitary-adrenal,HPA)軸發(fā)育及功能的影響。方法新生雄性SD大鼠54只,6日齡,隨機(jī)分為三組:對(duì)照組(C組)、七氟醚組(S組)和布美他尼+七氟醚組(BS組),每組18只。C組:吸入100%氧氣6h;S組:吸入2.1%七氟醚6h;BS組:吸入2.1%七氟醚6h,在麻醉前30min和麻醉后3h注射布美他尼。6h后,每組隨機(jī)選取6只處死收集血清,采用ELISA法測(cè)定皮質(zhì)酮濃度;各組剩余大鼠于第60天行高架十字迷宮測(cè)試,第65天行束縛應(yīng)激30 min,并在應(yīng)激前后取尾靜脈血,采用ELISA法測(cè)定血清皮質(zhì)酮濃度。結(jié)果麻醉后S組血清皮質(zhì)酮濃度明顯高于C組(P0.01);BS組血清皮質(zhì)酮濃度明顯低于S組(P0.01)。S組進(jìn)入開(kāi)放臂次數(shù)明顯少于C組(P0.05),開(kāi)放臂停留時(shí)間明顯短于C組(P0.05),應(yīng)激前后血清皮質(zhì)酮濃度明顯高于C組(P0.05或P0.01);BS組進(jìn)入開(kāi)放臂次數(shù)明顯多于S組,開(kāi)放臂停留時(shí)間明顯長(zhǎng)于S組(P0.05),應(yīng)激前后血清皮質(zhì)酮濃度明顯低于S組(P0.05)。結(jié)論新生期七氟醚麻醉可導(dǎo)致HPA軸功能亢進(jìn),其機(jī)制與GABAA受體相關(guān)。
[Abstract]:Objective to study the effect of sevoflurane exposure on hypothalamic-pituitary-adrenal (Hypothalamic-pituitary-adrenal). Methods 54 male SD rats were randomly divided into three groups: control group (C group). Sevoflurane group (n = 18) and bumetanil sevoflurane group (n = 18) and bumetanil sevoflurane group (n = 18). Group C: inhaled oxygen for 100% hours; Group S: inhaled 2.1% sevoflurane for 6 hours; BS group: after inhaling 2.1% sevoflurane for 6 hours, bumetanil was injected 30 minutes before anesthesia and 3 hours after anesthesia. 6 rats in each group were randomly killed to collect serum. The concentration of corticosterone was determined by ELISA. The remaining rats in each group were tested by elevated cross maze on the 60th day and 30 mins by restraint stress on the 65th day, and the blood of the tail vein was taken before and after the stress. Results the concentration of corticosterone in group S was significantly higher than that in group C (P 0.01) after anesthesia. The serum corticosterone concentration in BS group was significantly lower than that in group S (P 0.01). The number of open arm entry in group S was significantly lower than that in group C (P 0.05), and the retention time of open arm was significantly shorter than that in group C (P 0.05). The concentration of corticosterone in serum before and after stress was significantly higher than that in group C (P0.05 or P0.01). The number of open arm entry in BS group was significantly higher than that in S group, and the open arm stay time was significantly longer than that in S group (P0.05). Conclusion sevoflurane anaesthesia in newborn stage can lead to hyperfunction of HPA axis, and its mechanism is related to GABAA receptor.
