一種強(qiáng)化戊二醛殺滅微生物效果與安全性的研究

【摘要】: 戊二醛被公認(rèn)為一種廣譜、高效、低刺激、低腐蝕、安全低毒、穩(wěn)定的消毒劑,作為一種冷消毒滅菌劑,被廣泛用于怕高溫、怕腐蝕的醫(yī)療器械的首選消毒劑。近年來,戊二醛類消毒劑的生產(chǎn)廠家逐年增加,劑型種類繁多,產(chǎn)品質(zhì)量參差不齊。我們在實(shí)際工作中發(fā)現(xiàn),單純根據(jù)產(chǎn)品使用說明書和《消毒技術(shù)規(guī)范》要求來配制和使用,無法最大限度地利用戊二醛的殺滅效果,不能完全反映戊二醛消毒劑的特點(diǎn),不利于指導(dǎo)應(yīng)用單位合理有效地使用戊二醛消毒劑。因此,在實(shí)驗(yàn)室對(duì)一種強(qiáng)化戊二醛——潔億牌戊二醛消毒液進(jìn)行了殺菌效果的系統(tǒng)分析,通過不同配方、不同影響因素下戊二醛含量、殺菌效果變化規(guī)律,從多方面評(píng)價(jià)該消毒劑的質(zhì)量,探討如何提高戊二醛消毒劑的殺菌效果,指導(dǎo)其實(shí)際應(yīng)用;同時(shí)也借此工作為戊二醛消毒劑配方的研究提供實(shí)驗(yàn)室依據(jù)。 為了解潔億牌戊二醛消毒液的殺菌性能、穩(wěn)定性及其影響因素,我們在實(shí)驗(yàn)室對(duì)其進(jìn)行了戊二醛含量測定、懸液定量殺滅試驗(yàn)、載體定性殺滅試驗(yàn)、殺菌影響因素測定、加速穩(wěn)定性試驗(yàn)、能量試驗(yàn);并通過模擬現(xiàn)場消毒滅菌試驗(yàn)、連續(xù)使用穩(wěn)定試驗(yàn)等驗(yàn)證了該消毒劑的實(shí)用消毒效果;通過金屬腐蝕性試驗(yàn)、急性經(jīng)口毒性試驗(yàn)、一次完整皮膚刺激試驗(yàn),了解該消毒劑對(duì)金屬的破壞性及對(duì)人的急性毒性。 研究結(jié)果如下: 1.潔億牌戊二醛消毒液為三元包裝,A劑為添加有0.25%聚氧乙烯脂肪醇醚的戊二醛,B劑為防銹劑亞硝酸鈉,C劑為緩沖劑碳酸氫鈉。使用時(shí)取5g/L的B劑和0.4g/L的C劑加入戊二醛溶液中。防銹劑亞硝酸鈉的添加會(huì)降低戊二醛的含量。在不同pH值下戊二醛含量的測定值有差異,隨著pH的升高而降低。 2.潔億牌戊二醛消毒液屬穩(wěn)定類消毒劑,具有高效、快速的殺菌特點(diǎn)。19200mg/L潔億牌戊二醛消毒液對(duì)細(xì)菌芽胞作用2h;480mg/L對(duì)細(xì)菌繁殖體作用10min、960mg/L作用5min或1920mg/L作用2min,殺滅對(duì)數(shù)值均≥5.00;1920mg/L潔億牌戊二醛消毒液對(duì)白色念珠菌作用15min,其殺滅對(duì)數(shù)值≥4.00,達(dá)消毒合格要求。 3.潔億牌戊二醛消毒液可用于多次消毒污穢物品,其實(shí)用參考劑量為600mg/L。
[Abstract]:Glutaraldehyde is recognized as a kind of disinfectant with broad spectrum, high efficiency, low irritation, low corrosion, safety, low toxicity and stability. As a cold sterilizing agent, glutaraldehyde is widely used as the preferred disinfectant for medical devices afraid of high temperature and corrosion. In recent years, the manufacturers of glutaraldehyde disinfectants have increased year by year. In our practical work, we found that the preparation and use of glutaraldehyde could not be used to the maximum extent and could not reflect the characteristics of glutaraldehyde disinfectant solely according to the instructions for the use of the product and the requirements of the Technical Specification for Disinfection. It is unfavorable to guide the application units to use glutaraldehyde disinfectant reasonably and effectively. Therefore, the bactericidal effect of a kind of Glutaraldehyde fortified glutaraldehyde disinfectant solution was systematically analyzed in the laboratory. The content of glutaraldehyde and the change of germicidal effect were determined by different formulations and different influencing factors. The quality of the disinfectant was evaluated from many aspects, and how to improve the germicidal effect of glutaraldehyde disinfectant was discussed to guide its practical application. It also provides laboratory basis for the formulation of glutaraldehyde disinfectant. In order to understand the germicidal performance, stability and influencing factors of Jie-yi brand glutaraldehyde disinfectant solution, we carried out the determination of glutaraldehyde content, suspension quantitative killing test, carrier qualitative killing test and germicidal influence factor in laboratory. Accelerated stability test, energy test; The practical disinfection effect of the disinfectant was verified by simulating field disinfection sterilization test and continuous use stability test. Through metal corrosion test, acute oral toxicity test and a complete skin irritation test, it is understood that the disinfectant is destructive to metals and acute toxicity to human. The results are as follows: 1. Jieyi brand glutaraldehyde disinfectant was ternary packed, A was glutaraldehyde with 0.25% polyoxyethylene aliphatic alcohol ether, B was rust inhibitor sodium nitrite, and C was buffer sodium bicarbonate. When used, 5g/L B and 0.4g/L C were added to glutaraldehyde solution. The addition of sodium nitrite will reduce the content of glutaraldehyde. There were differences in glutaraldehyde content at different pH values, which decreased with the increase of pH. 2. Jieyi brand glutaraldehyde disinfectant is a stable disinfectant with high efficiency and fast bactericidal characteristics. 19200mg/L Jieyi brand glutaraldehyde disinfection solution acts on bacterial spores for 2 h. The killing logarithm of 480mg/L on bacterial propagator was more than 5.00 when 5min or 1920mg/L were exposed to 5min or 1920mg/L for 2 min. When 1920mg/L Jiayi brand glutaraldehyde disinfectant was exposed to Candida albicans for 15 min, the logarithmic value of killing was greater than 4.00, which reached the standard of disinfection. 3. Jieyi brand glutaraldehyde disinfectant solution can be used to disinfect contaminated articles for many times, and its practical reference dose is 600 mg / L.
