miR-144在人牙周膜成纖維細胞中調(diào)控Toll樣受體2及骨唾液酸蛋白的表達

目的檢測與Toll樣受體2(Toll-like receptor 2,TLR2)呈負(fù)相關(guān)性的微小RNA(microRNA,miRNA)在人牙周膜成纖維細胞(human periodontal ligament fibroblasts,hPDLFs)中與TLR2的作用關(guān)系及其對骨唾液酸蛋白(bone sialoprotein,BSP)的調(diào)控作用。方法查找與TLR2具有靶向結(jié)合關(guān)系且呈負(fù)相關(guān)性的miRNA(miR-23a、miR-144、miR-19a), Real-time PCR方法檢測正常及牙齦卟啉單胞菌(Porphyromonas gingivals,P.gingivalis)胞壁成分脂多糖(lipopolysaccharide,LPS)誘導(dǎo)下hPDLFs中目標(biāo)miRNA的mRNA表達水平;分空白對照組、陰性對照(LPS)組、miRNA過表達對照組、miRNA過表達組、miRNA抑制對照組、miRNA抑制組,用Lipofectamine 2000分別轉(zhuǎn)染hPDLFs,Real-time PCR及Western Blot法分別測定TLR2、BSP的mRNA及蛋白表達水平。結(jié)果經(jīng)LPS...
