局部注射黃芪多糖對大鼠正畸牙保持階段BMP-2表達的影響

【摘要】:目的:探討局部注射黃芪多糖對大鼠正畸牙保持階段牙周組織改建和骨形成蛋白2(BMP-2)表達的影響。方法:48只8周齡雄性Wistar大鼠隨機分為8組,每組6只,即空白對照組(1組),陰性對照組(1組),實驗組(6組)。利用50g牽引力近中移動右側上頜第一磨牙,建立大鼠正畸牙移動模型,加力21d。實驗組于拆除裝置前1d開始將10mg/L黃芪多糖溶液局部注射于右上第一磨牙遠中頰側黏骨膜下,每1次/3d,每次50μL;陰性對照組每3d注射等量生理鹽水。分別于1、3、7、14、21、28d后處死,測量正畸牙復發(fā)距離,采用HE染色觀察牙周組織變化,免疫組化染色檢測牙周組織中BMP-2的表達。結果:實驗組牙周膜寬度保持較好,除1d組外,相同時間的實驗組復發(fā)距離小于對照組,差異有統(tǒng)計學意義(P0.05)。在整個保持階段,對照組遠中側BMP-2表達強度逐漸減少,實驗組遠中側BMP-2表達強度逐漸增大。除1d組外,相同時間的實驗組BMP-2表達強度顯著高于對照組,差異有統(tǒng)計學意義(P0.05)。結論:大鼠正畸牙保持階段,局部注射黃芪多糖能夠增加牙周組織中BMP-2的表達,從而加速牙槽骨的改建,有利于正畸后牙齒的保持。
[Abstract]:Aim: to investigate the effect of local injection of astragalus polysaccharide on periodontal tissue remodeling and bone morphogenetic protein 2 (BMP-2) expression during orthodontic tooth retention in rats. Methods: 48 8-week-old male Wistar rats were randomly divided into 8 groups with 6 rats in each group, namely, blank control group (group 1), negative control group (group 1) and experimental group (group 6). The orthodontic tooth movement model of rats was established by using 50g traction force to move the right maxillary first molar near the middle, and the force was increased for 21 days. The experimental group began to inject 10mg/L astragalus polysaccharide solution into the distal buccal mucoperiosteum of the right upper first molar one day before the device was removed, each time every 3 days, 50 渭 L each time, and the negative control group was injected with the same amount of normal saline every 3 days. After 1, 3, 7, 14, 21, 28 days, the recurrence distance of orthodontic teeth was measured, the changes of periodontal tissue were observed by HE staining, and the expression of BMP-2 in periodontal tissue was detected by immunohistochemical staining. Results: the width of periodontal ligament in the experimental group was better than that in the control group, except for 1 day group, the recurrence distance of the experimental group was smaller than that of the control group at the same time, and the difference was statistically significant (P 0.05). During the whole retention stage, the expression intensity of BMP-2 in the control group decreased gradually, while the expression intensity of BMP-2 in the experimental group increased gradually. Except for 1 day group, the expression intensity of BMP-2 in the experimental group was significantly higher than that in the control group at the same time, the difference was statistically significant (P 0.05). Conclusion: in orthodontic tooth retention stage, local injection of astragalus polysaccharide can increase the expression of BMP-2 in periodontal tissue, thus accelerate alveolar bone remodeling, which is beneficial to tooth maintenance after orthodontic treatment.
